摘 要:SD 大鼠60只,平均体重195g,分为对照组(Ec)、维生素 E100mg(E_1)、150mg(E_2)三组,E_1,E_2均在试前1h 灌胃。乌拉坦腹腔麻醉,人工呼吸机维持呼吸。结扎左冠状动脉(LCA)5min 后开放。在结扎后即刻、2、5min;开放后即刻、2、5、15、 0、60min 同步记录心电图、动脉压、左室压、dp/dt 及心外膜电图。60min 后取左室心肌标本作电镜检查。结果:心电图、心外膜电图 ST 段均抬高;开放后 E_1、E_2的左室压能维持80~90mmHg 水平约30min,Ec 则继续下降。动脉压、dp/dt 的变化与左室压一致。Ec 的室性心律失常发生率、室颤发生率、死亡率、60min 存活率分别为95%、45%、55%、40%,而 E_1、E_2分别为35%、15%、10%、90%;55%、30%、15%、85%。电镜下 Ec 的心肌纤维断裂,破坏;细胞膜、线粒体膜破坏,线粒体肿胀,嵴消失。E_1、E_2的细胞膜、线粒体膜、溶酶体膜、闰盘均完整。表明维生素 E 可减少心律失常,抗心律失常,保持细胞膜完整,维持心脏功能,降低死亡率,延长生存时间。剂量100mg似略优于150mg。Sixty rats(SD)of either sex,weighing 150 to 280g were divided into control (Ec),vitamin E 100mg(E_1)and 150mg(E_2)groups.All rats were anesthetized with urethane.The respiration was maintained using an animal respirator.After heparin was administered,two catheters were inserted into both left and right carotid arteries for measurement of arterial and left ventricular pressure.The left coronary artery(LCA) was reversibly ligated and after 5 minutes the suture was removed and reperfusion was performed for 60 minutes.ECG,arterial pressure,left ventricular pressure,dp/dt and epicardial mapping were recorded at intervals of 2,5,15,30,and 60 min after li- gation and opening of the LCA,then the muscle samples were taken from left ventri- cle(LV) for electron microscopic examination.ST segments in both ECG and epicardial mapping were elevated.When the LCA was open,the LV pressure in the rats from E_1 and E_2 rose and was maintained at the level of 80—90mmHg for 30 min,but that in the rats from Ec decreased continuously.The arterial pressure and dp/dt are in the same way.The morbidity rate from ventrieular arrhythmia and ventricular fibrillation, mortality rate and 60-minute survival rate in the rats from Ec are 95%,45%,55%, and 40%,respectively,in the rats from E_1 and E_2 35%,15%,10%,and 90% and 55%,30%,15%,and 85%,respectively.Electron microscopic examination showed that the muscle fibre,membrane of muscle,mitochondria were ruptured and destroyed (Ec),but in rats from E_1 and E_2,the membrane of muscle cells,mitochondria,in- tercalated disc and lysosome remained intact.Vitamin E was suggested to be effective in decreasing and against ventricular arrhythmia,supporting ventricular function,ma- intaining the intactness of cell membrane,decreasing the mortality rate and increasing survival rate.
