摘  要:目的:旨在探讨双基因(VP3、IL-18)抑制骨肉瘤S-180的联合抑瘤作用。方法:将VP3、IL-18基因插入pVAX1表达载体的pCMV启动子之下的EcoRⅠ位点,构建成pVVP3及pVVP3IL-18。将该重组质粒与单独表达凋亡素基因的重组质粒分别注射荷S-180肿瘤细胞的BALB/C小鼠,按照质粒与脂质体3:1的比例混合,每3天注射1次,连续注射3次,比较抑瘤趋势和抑瘤率。结果:联合应用VP3基因和IL-18基因的重组病毒治疗组对肿瘤的抑瘤效果明显优于单独应用VP3基因的重组病毒治疗组。结论:凋亡素基因具有凋亡诱导效应及体内的抑瘤效应。凋亡素基因VP3与IL-18基因有协同抑瘤效应,VP3、IL-18联合基因治疗骨肉瘤可显著提高抑瘤效果。Objective:To investigate the antitumor efficacy of VP3 and IL-18 in human osteosarcoma S-180 cells.Methods:Eukaryotic expression plasmid pVVP3,pVVP3 and IL18 were constructed by inserting apoptin gene and IL-18 gene into the downstream of CMV promotor.The S-180 melanoma model in BALB/c was constructed and the mice were injected intratumorly with pVVP3,pVVP3 and IL-18 mixed with liposome for 3 times,and each with 100μg recombinant plasmid.The rate of tumor suppression after 3 times of injection was measured.Results:It was suggested that the combinative application of VP3 and IL-18 was better in resulting in tumor suppression then VP3.Conclusion:The combination of VP3 gene and IL18 gene has a potent antitumor efficacy.
