摘 要:目的 研究赖氨匹林(Aspisol)对人恶性黑色素瘤A375细胞凋亡的诱导作用。方法采用四甲荃偶氮唑蓝(MTr)比色法检测赖氨匹林对A375细胞增殖的抑制作用,倒置显微镜下观察A375细胞形态学变化,流式细胞仪(FCM)检测不同浓度的赖氨匹林诱导A375细胞的早期凋亡率。结果不同浓度的赖氨匹林对A375细胞均有明显的生长抑制作用,且其作用具有时间和浓度依赖性,差异具有非常显著性(P〈0.01)。不同浓度的赖氨匹林对A375细胞作用24h后,均可诱导其发生早期凋亡,并且呈浓度依赖性,差异具有显著性(P〈0.05)。结论赖氨匹林能抑制A375的增殖,诱导其发生凋亡。Objective To study the apoptotic induction of Aspisol on melanoma A375 cells. Methods The growth inhibition effect of Aspisol on A375 cells was evaluated by MTT assay. Morphological assay was observed by inverted microscope. Early apoptotic rate in A375 cells was analyzed by flow eytometry. Results The results of MTr assay demonstrated that the growth of A375 cells was inhibited by Aspisol in a time-and concentration-dependence (P 〈 0. 01 ). FCM analysis showed that Aspisol could markedly induce apoptosis ; the early apoptotic rate of A375 ceils exposed to different concentrations of Aspisol for 24 h were significant differences ( P 〈 0.05 ) compared with control group. Conclusion Aspisol could inhibit proliferation and induce apoptosis of A375 cells.
