试剂ZS预防和治疗大鼠亚硒酸钠诱导白内障的研究

摘  要:目的:探讨试剂ZS对亚硒酸钠诱导大鼠白内障的预防和治疗作用。方法:(1)预防实验:将未睁眼的8~10d龄Wistar大鼠26只,随机分为3组,即预防组、病理对照组和正常对照组。正常对照组腹腔注射生理盐水(0.1ml/10g),1次/d;病理对照组,腹腔注射亚硒酸钠溶液(30μmol/kg)一次造模后,30min内再腹腔注射生理盐水(0.1ml/10g),其后按同样剂量腹腔注射生理盐水,1次/d;预防组按同样方法一次造模后,30min内首次腹腔注射试剂ZS(20μmol/kg),其后按相同剂量腹腔注射试剂ZS,1次/d,所有动物共注射6d。在实验第7d对所有实验动物用裂隙灯检查晶状体混浊情况,然后测定晶状体匀浆中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;(2)治疗实验:将造模成功的37只动物分为病理对照组和治疗实验组(1个月和3个月),在不同时间对各组动物用裂隙灯检查晶状体混浊情况,测定晶状体匀浆中SOD活性和MDA含量。结果:(1)预防实验:预防组晶状体混浊程度低于病理对照组(P<0.01),而高于正常对照组(P<0.01);预防组晶状体中的SOD活性高于病理对照组(P<0.01),而低于正常对照组(P<0.01);预防组晶状体中的MDA含量低于病理对照组(P<0.01),而高于正常对照组(P<0.01)。(2)治疗实验:各治疗组晶状体混浊程度、晶状体中SOD活性和MDA含量与病理对照组比较无明显差异(P>0.1)。结论:试剂ZS对Wistar大鼠经亚硒酸钠诱导的白内障有显著的预防作用,但对Wistar大鼠经亚硒酸钠诱导已经形成的白内障无明显的治疗作用。Objective:To observe the preventive and curative effects of agent ZS on selenite - induced cataractous lenses in rats. Methods: 1. Preventive effects : 26 Wistar rat offsprings ( 8 - 10 days of age) were divided into three groups: normal control, model ( pathological control) and protection group. The animals in both model and protection group undergone sodium selenite intraperitoneal injection at 30 μmol/kg and then the animals in protection group received agent ZS intraperitoneal injection (20 μmol/kg) within 30 min following the same dose of agent ZS injection once a day,while those in model group only received normal saline intraperitoneal injection (0.1 ml/10 g) within 30 rain following the same dose of normal saline injection once a day. However, the normal control group was only given normal saline (0. 1 ml/10 g) intraperitoneal injection once a day. On day 7, the transparency of all lenses of all animals were observed under slit lamp microscopy. At the same time, the superexide dismutase (SOD) activity and malonaldehyde (MDA) level in lens homogenate were measured, respectively. 2. Therapeutical effects: 37 rats with selenite - induced cataract were divided into three groups : model group, ZS treated for 1 month group and treated for 3 months group. Two ZS treated groups were divided into three subgroups: eyedrop application,intraperitoneal injection low dose and high dose group. After treatment for 1 and 3 months, the transparency of the rat lenses was observed under slit lamp microscopy, respectively. At the same time, the SOD activity and MDA level in lens homogenate were measured. Results : In preventive effect experiment, the lens opacity in protection group was slighter than that in model group, but more severe than that in normal control group ( P 〈 0.01 ) ; The SOD activity in protective group was higher than that in model group, hut lower than that in normal control group (P 〈 0.01 ) ;The MDA level in protective group was lower than that i
