摘 要:目的:研究X-射线对肺癌细胞X线修复交叉互补基因1(X-ray repair cross complementing gene 1,XRCC1)表达的影响,探讨DNA碱基切除修复机制在肺癌细胞对放疗耐受中的可能作用。方法:以噻唑蓝还原法(MTT)检测X-射线对肺癌细胞(人肺腺癌细胞株A549)抑制率的影响,实时荧光定量PCR技术检测X-射线处理后肺癌细胞XRCC1 mRNA的表达水平。结果:肺癌细胞抑制率多数情况下随X-射线照射时间的延长及照射剂量的增大而增加,呈照射时间依赖性(P<0.05)和剂量依赖性(P<0.05);X-射线照射后肺癌细胞XRCC1 mRNA的表达水平均先增高后降低,4 Gy组和8 Gy组在72 h表达水平最高(P<0.05),16 Gy组和32 Gy组在48 h表达水平最高(P<0.05)。结论:X-射线照射可引起肺癌细胞XRCC1 mRNA表达水平的明显改变,提示DNA碱基切除修复机制可能在肺癌细胞对放疗的耐受中起了重要的作用。Objective: To study the effect of X-ray on expression levels of X-ray repair cross-complementing gene 1 (XRCC1) in lung cancer cells, and explore the effect of DNA base-excision repair mechanism in the resistance to radiotherapy of lung cancer cell. Methods : Cell inhibition ratio was measured using MTF assay. The expression level of XRCC1 mRNA was measured by RFQ-PCR assay. Results: In the majority, the rate of proliferation inhibiting of lung cancer cells was increased along with the radiation time of X-ray prolonging ( P 〈 0.05 ) and the increase of radiation dose ( P 〈 0.05 ). The expression level of XRCC 1 mRNA in the lung cancer cells was increased significantly after treatment with X-ray, and then decreased. The highest expression levels of XRCC1 mRNA of 4 Gy group and 8 Gy group were at 72 h(P 〈0.05), and the highest expression levels of XRCC1 mRNA of 16 Gy group and 32 Gy group were at 48 h(P 〈0.05) after treatment with X-ray. Conclusion : X-ray had a fairly obvious function to decreases proliferation of lung cancer cell, and it could induce the increase of the expression levels of XRCC1 mRNA in lung cancer ceils. The results suggest that DNA base-excision repair mechanism may play an important role in resistance to radiotherapy of lung cancer cell.
