摘 要:目的:通过给NOD/SCID小鼠尾静脉注射白血病人骨髓单个核细胞(BMMNC)或CEM细胞株的方式,建立人白血病-NOD/SCID小鼠髓外浸润模型,并进行模型鉴定。方法:将36只NOD/SCID小鼠经137Cs全身照射270cGy后,随机分为4组,每组9只,24h内尾静脉注射接种白血病人BMMNC或CEM细胞系。实验Ⅰ组:BMMNC1×107/只;实验Ⅱ组:BMMNC2×107/只;实验Ⅲ组:CEM细胞1×107/只;对照组:生理盐水0.2ml/只。持续观察其一般情况和生存时间,监测外周血白细胞计数和涂片,并应用组织病理、流式细胞术等监测白血病细胞髓外浸润表现。结果:注射4-8周后,三组实验组均发生髓外浸润,成功建立人白血病-NOD/SCID小鼠髓外浸润模型。实验Ⅰ组有5只成功建立模型;实验Ⅱ组7只成功建立模型;实验Ⅲ组9只均成功建立模型。结论:应用尾静脉注射白血病人BMMNC或CEM细胞株的方法,可成功建立人白血病-NOD/SCID小鼠髓外浸润模型,且尾静脉注射白血病人BMMNC成模率高,生存时间长,为白血病髓外浸润机制的研究和抗白血病髓外浸润药物的筛选提供了良好的实验模型。Objective:To build model of outer marrow infiltration of leukemia - NOD/SCID mice by injecting leu kemia patient's bone marrow mononuclear cells(BMMNC) and CEM cell line into tail vein. Methods: Mice ,irradiated 270cGy on body by ^137 Cs, were randomly divided into four groups, each group were 9. Inoculating leukemia patient's bone marrow mononuclear ceils(BMMNC) and CEM cell line within 24 hours, group Ⅰ :BMMNC 1 × 10^7 per mouse ;group Ⅱ :BMMNC 2 × 10^7 per mouse; group Ⅲ CEM cell 1 × 10^7 per mouse;control group: 0.2ml saline per mouse to observe the general situation and survival time, monitor peripheral blood white blood cell count and smear, outer marrow infiltration of leukemia cells by histopathology and flow cytometry. Results : 4 - 8W after the injection, all the experiment groups took place outer marrow infiltration , succeeded in building model. Group Ⅰ had 5 mouse which were made model successfully . Group Ⅱ were 7, while group m were 9. Conclusion : The model of outer marrow infiltration of leukemia - NOD/SCID mice can be made by in jecting leukemia patient's bone marrow mononuclear cells (BMMNC) and CEM cell line into tail vein of mice, and this method have higher making model success rate and longer survive time, provide a good model for study of mechanism of leukemia outer marrow infiltration and screening drug of anti - leukemia outer marrow infiltration.
