摘　　要：目的:探讨Celecoxib对重症急性胰腺炎环氧化酶-2(COX-2)表达的影响。方法:逆行胰胆管注射灭菌的4%牛磺胆酸钠诱导SD大鼠急性出血坏死性胰腺炎模型(AHNP)。SD大鼠随机分为假手术组、AHNP组及Celecoxib预处理组(C+AHNP),动态观察3、6、12h血清淀粉酶(AMS)及胰腺病理组织学改变,免疫组化SP法检测COX-2蛋白的表达。结果:AHNP组胰腺组织COX-2表达和病理学评分显著并进行性增加,Celecoxib预处理在各时相点均显著减轻胰腺损伤的程度,下调COX-2的表达水平(P<0.05)。结论:COX-2参与了急性胰腺炎的发生和发展,Celecoxib通过抑制COX-2的表达发挥对AHNP的保护作用。Objective：To investigate the influence of Celecoxib on cyclooxygenase-2（COX-2） expression in severe acute pan creatitis.Methods：The acute hemorrhagic and necrotic pancreatitis（AHNP） model was induced by standard retrograde infusion of bilio-pancreatic duct with 4% sterile sodium taurocholate solution in SD rats.Rats were randomly divided into sham group, AHNP group and C＋AHNP group.Enzyme chemical methods, HE and immunohistochemistrical staining were employed respectively to assess the alteration of AMS, histopathology and COX-2 expression of pancreatitis tissue over a time course of 3, 6 and 12 hours.Results：In the AHNP group, AMS and histopathology alteration and COX-2 expression of pancreatitis lung were lower than AHNP group at the same time（P〈0.05）.Conclusion：COX-2 plays an important role in severe acute pancreatitis, and Celecoxib can reduce inflammatory injury of pancreatitis in AHNP by down-regulating the expression of COX-2.
