摘 要:系统考察负载型尿素柱层析分离α-亚麻酸乙酯过程中残留甲醇量对分离效果的影响。结果表明:在尿素与硅胶载体比例为2∶3时,固定相中残留6%~12%的甲醇时对于饱和及单不饱和脂肪酸有较好的包合效果,可得到纯度超过80%的α-亚麻酸乙酯;残留甲醇量低于3%时,负载型尿素对于饱和脂肪酸乙酯的包合作用减弱甚至消失,反而是空间位阻小的饱和及单不饱和脂肪酸先洗脱;残留甲醇量高于12%时,随着时间的推移,负载型尿素对于含量较高的油酸乙酯包合作用逐渐减弱,导致洗脱液中α-亚麻酸乙酯含量降低。The effect of methanol residue on separation of ethyl α-linolenate by urea column chromatography was investigated. When the ratio of urea to silica gel is 2:3 (w/w), the methanol residue is 6%-12%, the ethyl esters of saturated fatty acids and monounsaturated fatty acids can be adducted well and the purity of ethyl α-linolenate is higher than 80%. When the methanol residue is lower than 3%, the adducting action to the ethyl esters of fatty acids by loaded urea becomes weaker even vanishes, even more, the ethyl esters of saturated fatty acids and monounsaturated fatty acids are eluted firstly. When the methanol residue is more than 12%, the adducting action to ethyl oleate is weakened, which induced the decrease of ethyl α-linolenate content in elutes.
