摘 要:应用光镊俘获单个红细胞并收集其拉曼光谱进行细胞识别,针对系统噪声和荧光效应等引起高强度光谱背景从而影响基于光谱强度统计分析结果的问题,利用最小二乘多项式进行光谱背景的拟合并去除背景。比较了4种数据处理方法(去背景、未去背景、去背景做一阶导数和未去背景做一阶导数)的聚类效果,结果表明,去背景的处理方法显著提高了地贫红细胞与正常人红细胞的识别度。其中,α地贫HbH组与正常对照组间的平均区分度为90.32%,α地贫HbHcs组与正常对照组间的区分度为97.6%,远高于其他处理方法的结果。同时,去背景及未去背景的一阶导数光谱的聚类分析结果一致,证实去背景并不影响光谱特征谱带的形状及其性质。此外,该方法对实验系统稳定性要求不高。Thalassemia results from inherited defects in the production of hemoglobin. In this paper, a new method for discriminating thalassemic erythrocyte types using Raman spectroscopy was reported. A laser tweezers was used to trap a single erythrocyte which was then studied by Raman spectroscopy. Blood samples collected from alpha-thalassemia patients with HbH and HbHcs disease, and normal persons were used for examination. A least-square polynomial fitting was proposed to eliminate the high-intensity background of spectral signal caused by system noise and fluorescence which would otherwise seriously interfered the results of statistical analysis. Four different kinds of Raman spectra, namely the raw spectra, the background-removed spectra, the first derivative raw spectra and the first derivative background-removed spectra, obtained from the corresponding patients and normal persons were processed by the cluster analysis respectively. The results indicated that the discrimination be- tween the erythrocytes obtained from patients and normal person was significantly improved by using the background-removed spectra. The discrimination index between HbH and normal groups was 90.32% , and that between HbHcs and normal groups was 97.6% , which were higher than the results obtained from other spectra. In addition, the result of the first derivative background-removed spectra was very similar to that of the first derivative raw spectra, indicating that the removal of background did not affect the basic spectral shapes and characteristics. The result also showed that some variations of experimental conditions did not affect the analysis results. This study may provide a useful tool for discriminating and diagnosis of thalassemic erythrocytes.
