摘  要:目的:通过观察消痰散结方对裸鼠人胃癌MKN-45原位移植模型端粒酶逆转录酶(hTERT)蛋白及mRNA表达的影响,探讨其抑制胃癌细胞增殖的作用机制。方法:采用OB胶粘贴法建立裸鼠人胃癌MKN-45原位移植瘤模型,随机分成模型组、消痰散结方低剂量组、消痰散结方中剂量组、消痰散结方高剂量组、5-Fu组,每组8只,运用免疫组化、实时荧光定量RT-PCR方法检测hTERT蛋白及mRNA的表达。结果:①消痰散结方低、中、高剂量组的平均瘤重与模型组相比,有显著差异(P<0.01);②消痰散结方中、高剂量组hTERT蛋白的阳性表达率与模型组相比有统计学意义(P<0.05或P<0.01);③消痰散结方中、高剂量组hTERTmRNA含量与模型组相比有统计学意义(P<0.05)。结论:①消痰散结方有明显的抑制胃癌细胞生长的作用;②消痰散结方通过下调hTERT蛋白及mRNA的表达而抑制端粒酶的活性,使癌细胞端粒长度逐渐缩短,丧失无限增殖能力,起到抑制肿瘤细胞生长、增殖的作用。Objective:To explore the mechanism of Xiaotan Sanjie Fang( XTF), a traditional Chinese medicinal empirical formula for curing gastric cancer,in inhibiting proliferation of gastric cancer cells by observing its influence on the expression of human gastric cancer MKN-45 telomerase reverse transcriptase (hTERT) and mRNA in a nude mice in situ tumor transplantation model. Methods:Using OB glue,the nude mice human gastric cancer MKN-45in situ transplantation tumor model was established, based on which 40 animals were equally randomized into 5 groups : the blank model group, low-dose XTF group, intermediate-dose XTF group, high-dose XTF group, and 5-Fu group, hTERT protein and mRNA expressions were assayed by immunohistoehemistry and RT-PCR. Results : 1 ) There was a significant difference in mean tumor weight between the low- , intermediate- , and high-dose XTF groups and the model group(P〈0. 01 ) . 2)There was a significant difference in hTERT positive expression between the intermediate- and high-dose XTF groups and the model group (P 〈 0. 05 or P 〈 0. 01 ). 3 )There was a significant difference in hTERT mRNA content between the intermediate- and high-dose XTF groups and the model group( P 〈 0. 05 or P 〈 0. 01 ). Conclusion : 1 ) XTF has a marked inhibitory effect on the growth of gastric cancer cells. 2)XTF inhibits hTERT by down-regulating expression of hTERT protein and mRNA, thus shortening the length of telomeres gradually, rendering it unable to proliferate indefinitely and finally inhibiting the grnwth nnd proliferntion of tumor cells.
