软脉煎对家兔颈动脉粥样硬化模型血管平滑肌细胞凋亡的影响

摘　　要：目的:通过对颈动脉粥样硬化家兔模型的治疗观察,研究软脉煎对动脉粥样硬化形成晚期血管平滑肌细胞凋亡影响。方法:采用高脂喂养加颈动脉内膜空气干燥术制备家兔颈动脉粥样硬化模型,38只新西兰家兔随机分成4组,正常对照组8只,模型组8只,软脉煎组11只,辛伐他汀组11只,12周后光镜观察颈动脉的形态并测量动脉内膜及中膜厚度并采用TUNEL法检测颈动脉血管平滑肌细胞的凋亡情况。结果:软脉煎组与模型组比较,能减轻颈动脉内膜增厚,具有显著性意义(P<0.05),能降低颈动脉血管平滑肌细胞的整体凋亡率及减少斑块肩部和纤维帽处的平滑肌细胞凋亡,均有显著性意义(P<0.05)。结论:软脉煎对动脉粥样硬化有一定的防治作用,并能在动脉粥样硬化形成晚期维持斑块稳定性,其机制可能与减少血管平滑肌细胞凋亡有关。Objective ：Through the treatment on rabbit carotid atherosclerosis model with Ruanmaijian, observe the effection on apoptosis in vascular smooth muscle cells. Methods ： A rabbit carotid atherosclerosis model was established by the high-fat diet and air drying method. 38 New Zealand rabbits were randomly divided into four groups, and normal control group eight, model group 8, Ruanmaijian Group 11, simvastatin group 11. Stablish the rabbit model of atberosclerosis. After 12 weeks ,optical microscopy carotid morphology measurement of intima-media thickness, and cells apoptosis in carotid artery vascular smooth muscle by TUNEL method was performed. Results ： It was found that the decoction effectively decreased carotid intima - media thickness （P 〈 0. 05 ） and late-stage atherosclerotic vascular smooth muscle cell apoptosis was inhibited （P 〈 0. 05 ）. Conclusion：Ruanmaijian have some control on atherosclerosis, and play a better role in maintaining stability of AS plaque. The mechanism may be the reduction of vascular smooth muscle cell apoptosis.
