摘 要:目的:探讨电针足三里穴对盲肠结扎穿孔(cecal ligation and puncture,CLP)致腹腔感染脓毒症大鼠脾淋巴细胞凋亡及相关蛋白Bc l-2表达的影响。方法:将40只SD大鼠随机分成正常对照组、模型组、非经非穴组、足三里组。采用CLP致腹腔感染脓毒症模型,应用流式细胞仪以Annexin-PI双染色法及光镜、透射电镜观察CLP致腹腔感染后36h脾淋巴细胞的凋亡情况,并观察脾淋巴细胞中Bc l-2蛋白含量的变化。结果:CLP致腹腔感染脓毒症可导致脾淋巴细胞凋亡增多,模型组、非经非穴组、足三里组的胸腺细胞凋亡率分别为(33.3%±5.1%)、(36.1%±5.3%)、(21.7%±4.6%),高于正常对照组(6.5%±2.1%)(P<0.05);电镜和光镜显示脾淋巴细胞凋亡典型的形态学特征;而足三里组的脾淋巴细胞凋亡率低于模型组和非经非穴组(P<0.05)。CLP致腹腔感染后脾淋巴细胞Bc l-2蛋白含量下降,模型组、非经非穴组、足三里组分别为(32.4±5.2)、(37.5±4.8)、(56.0±4.9),低于正常对照组(82.6±5.1)(P<0.05);而足三里组的脾淋巴细胞Bc l-2蛋白含量高于模型组和非经非穴组(P<0.05)。结论:电针足三里穴可以减轻CLP致腹腔感染大鼠脾淋巴细胞凋亡,其机制可能与促进Bc l-2基因的表达、提高脾淋巴细胞中Bc l-2蛋白含量有关。Objective:To study the influence of electro-acupuncture(EA)at Tsusan-li on the apoptosis of splenic lymphocytes and apoptosis-related protein Bcl-2 expression in rats with sepsis induced bv abdominal infection resulted from cecal ligation and puncture(CLP).Methods:Total of 40 Sprague-Dawley(SD)rats were randomly divided into four groups,including control group,model group,non-acupoint group and Tsusan-li group.The sepsis model is made from abdominal infection resulted from CLP.The apoptosis of splenic lymphocytes was observed under electron microscope and light microscope and apoptosis ratio of splenic lymphocytes was determined by Annexin-PI method with flow cytometry technique The content of Bcl-2 protein of splenic lymphocytes was determined.Results:Sepsis made from abdominal infection resulted from CLP can significantly increase the apoptosis of splenic lymphocytes and lead to the the typical histopathology changes of apoptosis of splenic lymphocytes under electron microscope and light microscope.Apoptosis rates of splenic lymphocytes in model group(33.39%±5.1%),non-acupoint group(36.1%±53%) and Tsusan-li group(21.7%±4.6%)are significantly higher than control group(6.5%±2.1%)(P〈0.05),and apoptosis rate of splenic lymphocytes in Tsusan-li group is significantly lower than those in model group and non-acupoint group(P〈0.05).Sepsis made from abdominal infection resulted from CLP also can reduce the content of Bcl-2 protein of splenic lymphocytes.The content of Bcl-2 protein of splenic lymphocytes in model group(32.4±5.2),non-acupoint group(37.5±4.8)and Tsusan-li group(56.0±4.9)are significantly lower than control group(82.6±5.1)(P〈0.05).And the content of Bcl-2 protem of splenic lymphocytes in Tsusan-li group is significantly higher than those in model group and non-acupoint group(P〈0.05).Conclusion:EA at Tsusan-li can significantly reduce the apoptosis of splenic lymphocytes in rats with sepsis and that may be closely associated
