摘  要:目的:探讨大鼠脑缺血后血脑屏障(blood-brain barrier,BBB)通透性的变化。方法:制做永久性大脑中动脉阻塞(permanent middle cerebral artery occlusion,pMCAO)模型,之后在缺血后的不同时间点,从大鼠股静脉注射2%的伊文思蓝(Evens Blue,EB),通过荧光显微镜及甲酰胺提取两种方法来评估BBB对EB的通透性。结果:随着缺血时间的增加,脑组织EB蓝染的范围逐渐增大,荧光光斑数量增加,强度增强。甲酰胺法显示,EB的渗出量在3h时即有增加,12h达到高峰以后逐渐降低,其测定结果与荧光显微镜下的观察结果较为一致。结论:脑缺血后BBB的通透性增高,并在48h内呈单相变化。另外还发现,EB法不宜用来比较缺血区与非缺血区间BBB通透性的差异。Objective : To study the alteration of blood brain barrier(BBB) leakage after cerebral ischemia. Methods : The permanent middle cerebral artery occlusion model was induced in Wistar rat Subsequently,2% solution of Evans blue was administered intravenously from vena femoralis in a dose of 4mL/kg at expected time after MCAO. Then the BBB leakage was evaluated with fluorescence microscope and the method of formamide. Results : With the prolonging of the ischemic duration, the scope of EB blue-staining in eerebral tissue increased gradually, and the number and area of fluoreseent faculae increased till 12h. The formamide-method resultimplied that the extravasation of EB inereased at 3h after MCAO,and it achieved the peak at 12h. Then it stepped down gradually till 48h after ischemia. Conclusion :The alteration of BBB leakage during the 48h after cerebral ischemia is in a monophase. And it may be not a Correct method to evaluate the distinction of BBB leakage between ischemic region and non - ischemic region by EB.
