比较不同中药方剂对兔动脉内膜损伤后血管平滑肌细胞增殖的影响

摘　　要：目的:比较化痰祛瘀、益气养阴、疏肝理气3种不同方剂对兔动脉内膜损伤后血管平滑肌细胞(VSMCs)增殖的影响。方法:将46只新西兰兔随机分为正常组、模型组、氟伐他汀对照组、化痰祛瘀组、益气养阴组、疏肝理气组,正常组6只,其余每组各8只。通过高脂饲料配合腹主动脉内膜剥脱术建立兔动脉内膜损伤模型。取腹主动脉下段,电镜观察血管平滑肌细胞形态;应用RT-PCR法测定原癌基因c-myc和血小板源性生长因子-BB(PDGF)水平;免疫组化法测定胰岛素样生长因子-1(IGF-1)水平。结果:(1)模型组PDGF、c-myc、IGF-1均显著高于空白组(P<0.05)。(2)氟伐他汀组、化痰祛瘀组、益气养阴组、疏肝理气组与模型组相比较PDGF、c-myc、IGF-1显著下降(P<0.05)。(3)化痰祛瘀组、益气养阴组与疏肝理气组比较PDGF、IGF-1显著下降(P<0.05),c-myc差异不显著(P>0.05)。结论:(1)动脉损伤后可致PDGF、c-myc、IGF-1表达增加,提示PDGF、c-myc、IGF-1表达增加在动脉损伤后平滑肌细胞增殖中起一定作用。(2)化痰祛瘀、益气养阴、疏肝理气3种方剂均能使动脉损伤后PDGF、c-myc、IGF-1表达减少,对动脉损伤后平滑肌细胞增殖有一定抑制作用,化痰祛瘀与益气养阴方效果更显著。Objective ： To compare the effect of different traditional Chinese medicine such as Huatan Quyu, Yiqi Yangyin and Shugan Liqi on vascular smooth muscle cells（VSMCs） proliferation in rabbits after endarterium injury. Methods： Fourty-six New Zealand rabbits were randomly divided into normal group, model group, Huatan Quyu, Yiqi Yangyin and Shugan Liqi groups ,There were six rabbits in normal group and eight in others. Through feeding rabbits with high dosage of cholesterol diet along with endarterium denudation, rabbits endarterium-injured models were build. Vascular smooth muscle cells （VSMCs） were observed under the electron microscope. Reversetranseription-polymerase chain reation （ RT - PCR） method was used for observing the levels of c-myc and platelet derived growth factor- BB（ PDGF -BB）. The level of Insulin-like growth faetor-1 （ IGF - 1 ） was collected by immunohistochemistry. Results ： PDGF - B, e - myc and IGF - 1 increased significantly in modle group compared with the normal group （ P 〈 0. 05 ）. PDGF - B, c - myc and IGF - 1 decreased in three Chinese traditional medicine groups. PDGF - B and IGF - 1 decreased significantly in Huatan Quyu and Yiqi Yangyin groups compared with Shugan Liqi group（P 〈0. 05）. Conclusion：Mter artery injured,PDGF - B,c - mye,and IGF- 1 increased. Chinese traditional medicine can restrain smooth muscle cell proliferation and Huatan Quyu and Yiqi Yangyin were better than Shugan Liqi.
