摘  要:目的:建立人肺癌不同转移潜能细胞株,并对其转移特性进行初步研究,为肺癌转移机制的研究提供模型。方法:采用有限稀释单细胞克隆培养的方法,从已建立的人肺癌高转移细胞株SPC-A-1sci中分离出的不同单克隆细胞,通过体外损伤愈合、迁移与侵袭实验,体内皮下移植瘤自发性转移实验,比较分离出的不同细胞株之间以及与亲代高转移细胞株SPC-A-1sci之间运动、迁移、侵袭及转移能力的差异。结果:从SPC-A-1sci细胞株中成功分离出3株单克隆细胞(命名为SPC-A-1sci-C5、SPC-A-1sci-F3、SPC-A-1sci-C2),均具有特异性形态学特征;与亲代高转移细胞株SPC-A-1sci相比,SPC-A-1sci-C5细胞株的体外侵袭能力明显增强,其他2株细胞的体外侵袭能力则明显降低;3株单克隆细胞在细胞形态和体外侵袭能力方面存在明显差异,SPC-A-1sci-C5的体外侵袭能力明显高于其他2株细胞。体内皮下移植瘤自发性转移试验显示,SPC-A-1sci-C5细胞不仅转移发生时间较早,而且肺转移程度(包括转移灶数量及大小)亦较其他2株细胞增多。结论:成功建立了具有相同遗传背景、不同转移潜能的3株人肺癌细胞,为肺癌转移的进一步研究提供了理想模型。Objective : In order to provide the evidence to uncover the mechanisms for lung cancer metastasis, this study develops and characterizes a serial of human lung cancer cell strains with stepwise metastatic potentials. Methods : The different cell strains were isolated and cultured from a highly metastatic SPC-A-lsci cell line by using limited dilution and monoclonal method. The wound healing test and migration and invasion assays in vitro and spontaneous metastasis test in subcutaneously xenografted tumor in vivo were performed to compare the difference in the mobilization, migration, invasion, and metastatic potentials between isolated cell strains and its highly metastatic parent cell line (SPC-A-lsci). Results:Three different monoclonal cell strains (designated SPC-A-lsci-C5, SPC-A- lsci-F3, and SPC-A-lsci-C2) were isolated successfully from highly metastatic SPC-A-lsci cell line. The three cell lines had typical morphological features. Compared with parent SPC-A-lsci cells the invasion capability of SPC-A-lsci-C5 cells was markedly enhanced but the invasion capabilities of the other two cell lines were greatly weakened. There was a significant difference in the cell morphology and invasion capability between the three isolated cell lines. The invasion capability of SPC-A-lsci-C5 cells was stronger than that of the other two cell lines. Spontaneous metastasis test in subcutaneously xenografted tumor in vivo showed that the metastasis of SPC-A-lsci- C5 occurred early and the metastatic degree including the number and size of metastatic lesions were increased compared with the other two cell lines. Conclusion: This study successfully establishes three human lung cancer cell strains with stepwise metastatic potentials, which has same genetic background but different biologic behaviors. The study provides a well-defined cell model for further investigation on the molecular mechanisms for lung cancer metastasis.
