摘  要:目的:探讨雌激素、孕激素以及三苯氧胺(tamoxifen,TAM)对雌激素受体(estrogen receptor,ER)阴性的人子宫内膜癌KLE细胞中组织激肽释放酶(kallikrein,KLK)4mRNA和蛋白表达的影响。方法:将不同浓度的17-β雌二醇(17-βestra-diol,17-βE2)、醋酸甲地孕酮(megestrol acetate,MPA)和TAM作用于KLE细胞,MTT法检测细胞的增殖活性,实时荧光定量PCR(real-time fluorogentic quantitative PCR,RFQ-PCR)和Western印迹法检测KLK4mRNA和蛋白的表达水平。结果:不同浓度的17-βE2对KLE细胞增殖均有促进作用,以10-10mol/L17-βE2作用最明显,与对照组相比差异有统计学意义(P<0.05);雌激素可以上调KLE细胞中KLK4mRNA和蛋白质的表达,10-10和10-8mol/L的17-βE2上调作用最明显,与对照组相比差异具有统计学意义(P<0.01);MPA可以下调KLE细胞中KLK4mRNA和蛋白质的表达,与对照组相比差异有统计学意义(P<0.01);TAM不影响KLE细胞中KLK4mRNA和蛋白质的表达(P>0.05)。结论:雌、孕激素可以调控ER阴性的人子宫内膜癌KLE细胞中KLK4的表达,提示雌激素可能通过非经典核内受体调控模式影响子宫内膜癌的发生发展。Objective : To investigate the effects of estrogen, progestogen, and tamoxifen (TAM) on the expressions of kallikrein ( KLK)4 mRNA and protein (hK4) in human endometrial carcinoma cell line KLE with negative expression of estrogen receptor (ER). Methods: Human endometrial cancer cell line KLE was incubated with 17-βestradiol( 17-βE2 ), megestrol acetate (MPA), and TAM at different concentrations for 72 h, respectively. KLK4 mRNA and protein were extracted and their expression levels were evaluated by quantitative RT-PCR and Western blotting, respectively. Results:Compared with the control group, KLK4 mRNA and protein expression levels were significantly increased by 17-βE2. The effect of 17-βE2 was more obvious at concentrations of 10^-10 and 10^-8 mol/L (P 〈 0.01 ). However, the levels of KLK4 mRNA and hK4 protein were downregulated by MPA compared with control group ( P 〈 0.01 ). TAM did not affect the expression levels of KLK4 mRNA and hK4 protein in KLE cells ( P 〉 0.05 ). Conclusion: Estrogen and progesto- gen regulate the expression levels of KLK4 mRNA and protein in ER-negative human endometrial carcinoma cell line KLE. The results indicate that estrogen influenced the generation and progression of endometrial carcinoma via non-typical nuclear receptor regulating pathway.
