摘 要:目的:对简单节杆菌TCCC11037发酵生产脱氢酶的培养基进行优化。方法:利用单因子试验筛选出最适碳源为葡萄糖,氮源为酵母膏。采用Plackett-Burman(P-B)方法筛选出对产酶有重要影响的因素,并采用响应面试验设计(RSM)对重要因素进行优化。结果:葡萄糖、酵母膏、KH2PO4的浓度是影响脱氢酶产生的重要因素。优化后的培养基组成为(%):葡萄糖1.21,酵母膏0.65,KH2PO40.24,玉米浆0.8;培养基初始pH值7.0,接种量5%,通气条件为装液量100mL/500mL。结论:优化后脱氢酶活力达到24.57μmol/(g·min),得到了明显的提高。Objective:Optimize the fermention conditions of dehydrogenase by Arthrobacter simplex TCCC11037. Method: By single-factor test, the optimum carbon source and nitrogen source were confirmed as glucose and yeast extract respectively. The important factors were selected by Plackett-Burman method, and response surface analysis was used to optimize these important factors. Result: The dehydrogenase activity was mainly influenced by concentration of glucose, yeast extract and KH2PO4. The optimal fermention medium components(%) were: glucose 1.21, yeast extract 0.65, KH2PO4 0.24, corn steep liquor 0.08, the optimum enzyme producing conditions were: initial pH of medium 7.0, inoculum volume 5%, liquid volume in flask 100mL/500mL. Conclusion: At optimum conditions, the dehydrogenase activity was significantly improved to 24.57μmol/(g·min).
