摘 要:目的探讨北虫草提取物对冷应激大鼠不同脑组织区域环磷酸腺苷(cAMP)水平与腺苷酸环化酶(adenylate cyclase,AC)活性的影响。方法采用4℃暴露3h应激方式,持续应激4周,建立大鼠冷应激模型,用酶联免疫吸附实验(ELISA)、放射免疫分析(RIA)法检测脑皮质,脑干和丘脑下部cAMP含量和AC活性的改变。结果冷应激实验组与正常对照组比较大鼠脑皮质、脑干,丘脑下部cAMP含量和AC活性明显下降,虫草提取物能明显拮抗冷应激引起的cAMP含量和AC活性降低。结论北虫草提取物对冷应激大鼠脑组织不同区域cAMP含量和AC活性有明显的改善作用,提示持续冷应激会导致中枢神经系统功能障碍。而虫草提取物表现出的上调cAMP含量和AC活性作用可能与增强冷应激大鼠脑组织不同区域交感神经-肾上腺-靶细胞信号传导网络之间的功能有关,确切的调控机理尚需进一步深入实验研究。Aim To Study the effects of purify substance from Cordyceps militaris on the activity of AC and cAMP levels in different brain tisusse regions of cold stress rats. Methods stress fashion was used by exposing 3h at 4℃, cold stress Wistar rat pattern was established on successive 4W, the cAMP content was determined by ELISA, RIA was used to measure the activity change of AC in cortex, brainstem and hypothalamus of rat brain. Results compare with control group, cold stress test group, cAMP content and activity of AC reduce in cortex,brainstem and hypothalamus of rat brain, P 〈 0.05 . Purify substance from Cordyceps militaris can obviously resist lower of cAMP content and activity of AC, compare with cold stress test group, P 〈 0.05. Conclusion This treatise had demonstrated that purify substance from Cordyceps militaris can obviously improve cAMP content and activity of AC in different brain tisusse regions of cold stress rats. This shown lasting cold stress will result in function obstruct of central nervous system. That purify substance from Cordyceps militaris can regulate up cAMP content and activity of AC may be strengthener function among sympathetic-adermal-target cell signal conduction net in in different brain tisusse regions of cold stress rats. The best indeedly adjust principle want been investigated deely.
