摘 要:目的探讨牛磺酸对链脲佐菌素诱导糖尿病(DM)大鼠视网膜谷氨酰胺合成酶(GS)表达的影响。方法 DM大鼠接受1%、5%牛磺酸处理2周、1个月、2个月后取视网膜,用RT-PCR、免疫组织荧光化学法检测牛磺酸对视网膜GS mRNA和蛋白表达的影响。结果 DM后1个月,视网膜中GS mRNA表达开始减弱,随病程延长下降显著。DM2个月时整个视网膜GS免疫染色明显变浅,尤以Muller细胞胞体、内网状层、节细胞层变化最明显。牛磺酸干预可以增强视网膜GS的表达。结论牛磺酸可以通过增加DM视网膜GS的表达,改善DM引起的谷氨酸兴奋毒性,从而保护视网膜。Objective To investigate the effects of taurine treatment on retinal glutamine synthetase (GS) expression in diabetic rats. Methods Diabetic rats were fed with 1% and 5 % taurine for 2 weeks, 1 month and 2 months. RT-PCR and immunohistofluorescence were employed to detect the expressions of GS mRNA and protein. Results From the first to the second month, the mRNA expression of GS gradually decreased in diabetic retina. The GS expressions in Mueller cell bodies,inner plexiform layer and ganglion cells were decreased at 2 month after DM. Taurine up-regulated GS mRNA and protein expression in retina cells. Conclusions Taurine can up-regulate the mRNA and protein expression of GS and restrain excitotoxicity of glutamate. These observations suggest that taurine may play a role in diabetic retina protection.
