转化生长因子-β_1在肠易激综合征大鼠模型中表达的研究

摘　　要：目的：观察转化生长因子（TGF）-β1在肠易激综合征（IBS）大鼠模型肠粘膜组织中的表达，初步探讨TGF-β1在IBS发病机制中的作用。方法：采用乙酸灌肠加束缚应激和单纯束缚应激方法制造IBS动物模型（IBS1、IBS2组），同时设立灌肠对照和正常对照组，行内脏敏感性评价后，用免疫组化的方法检测TGF-β1在IBS大鼠模型组和对照组肠粘膜组织中的表达情况。结果：IBS1、IBS2两模型组大鼠腹外斜肌收缩次数在不同扩张容量下均较正常对照纽明显增加（P〈0．05—0．01），组织学分析显示各组大鼠均无明显的炎症性表现。TGF-β1在各组肠粘膜组织均有表达，各组之间的差异无统计学意义（P〉0．05）。结论：TGF-β1作为一个抗炎性细胞因子在IBS发病机制中可能通过非直接的方式发挥作用。Objective： To observe the expression of transforming growth factor -β1 in rats model with irritable bowel syndrome and investigate the effect of TGF-β1 on antiinflammatory cytokine. Methods： Rats model of IBS by intracolonic instillation of acid and restraint stess and single restraint stess methords, while the positive and control group were designed, after the visceral sensitivity was detected in the rats of each group, the expression of TGF-β1 was detected by immunohistochemistry. Results ：The contraction frequency of the abdominal muscle in two rat model groups was significantly enhanced as compared with two control groups （ P 〈 0.05- 0.01 ）. There were no signs of inflammation in the rats of each group. The expression of TGF-β1 different position of each group was no significant difference （ P 〉 0.05）. Conclusions： TGF-β1 may be an important regulatory factor in IBS which acts as a antiinflammatory cytokine by indirect means.
