摘 要:目的:观察放射治疗对BALB/c小鼠肾癌抑制作用及对TNF-α表达的影响。方法:用Renca肾癌细胞制作BALB/c小鼠皮下移植瘤模型,放疗组小鼠在种瘤第12~16天,连续5d进行肿瘤局部放射,第28天处死小鼠,取肿瘤组织测量,分析治疗效果。体外培养2×106Renca肾癌细胞,经20GyX射线单次照射培养24h后,收获细胞。提取细胞和肿瘤组织蛋白,免疫印迹法检测治疗后肿瘤组织和细胞中TNF-α蛋白的表达水平。结果:放射治疗组有效抑制肾癌细胞在BALB/c小鼠体内生长,局部放疗明显增加了肿瘤组织内TNF-α表达水平,体外培养的Renca肾癌细胞经放射线处理后TNF-α表达明显上调。结论:放射线能够促进肿瘤细胞分泌TNF-α,并且与肿瘤局部产生的TNF-α表达上调密切相关。Objective: To observe the inhibitory effect of radiotherapy on renal carcinoma and expression of TNF-α in BALB/c mice. Methods: Renal carcinomabarring mice were treated with local fractionated tumor irradiation. The mice were euthanized on day 28. Cultured Renca cells were exposed to a single radiation dose of 20Gy after cultured for 24h. Expression of TNF-α protein in tumors and Renca cells was detected by Western blot. Results: Radiotherapy inhibited the growth of renal carcinoma in BALB/c mice. Local radiotherapy significantly increased the expression of TNF-α in tumor tissue. The expression of Renca cells cultured in vitro was significantly up-regulated after radiotherapy. Conclusion: X-rays can stimulate tumor tissue to secrete TNF-α and are correlated with the up-regulation of TNF-α expression in tumor tissue.
