MMP-9和TIMP-1在高危骨肉瘤中的表达及其诊断意义

摘　　要：目的研究高危骨肉瘤中MMP-9和TIMP-1的表达情况,探讨MMP-9/TIMP-1失衡在高危骨肉瘤中的诊断意义。方法对14例高危骨肉瘤、41例恶性程度低的骨肉瘤和I5例正常骨组织行MMP-9和TIMP-1免疫组织化学染色,并对染色情况进行分析。结果高危骨肉瘤组的MMP-9的表达率为100%,呈普遍过表达,明显高于低恶性骨肉瘤组的73.2%的阳性率(P<0.05),而高危骨肉瘤组的TIMP-1的阳性率明显低于低恶性骨肉瘤组(P<0.05)。高危骨肉瘤组的MMP-9/TIMP-1表达失衡率为42.8%,明显高于低恶性骨肉瘤组的12.2%(P<0.05)。结论MMP-9与TIMP-1可作为诊断骨肉瘤的辅助指标。Objective To investigate the expression of MMP-9 and TIMP- 1 in high risk osteosarcoma specimens and the diagnostic significance of unbanlance of MMP-9/TIMP-1. Methods The expression of MMP-9 and TIMP-1 were evaluated immuno-histo- chemically in 15 normal bone tissues, 14 high risk nsteosarcoma and 44 osteosarcoma specimens. Results The expression of MMP-9 in high risk osteosarcoma group were markeclly higher than those of osteosarcoma group;while the expression of TIMP-1 in high risk osteosarcoma group were markedly lower than those of osteosarcoma group. The anbanlance of MMP-9/TIMP-1 of high risk osteosarcoma group were markedly higher than those of osteosarcoma group. Conclusion 1. MMP-9 and TIMP-1 may act as a symbol of malignant phenotype of osteosarcoma.2.The unbaalance of MMP-9/TIMP-1 may become a diagnostic criterion of high risk osteosarcoma.
