摘  要:目的:观察阿司匹林(ASA)对鼻咽癌SUNE细胞系放射敏感性的影响,并探讨其可能机制。方法:用MTT法检测ASA对SUNE细胞的增殖抑制率,流式细胞仪检测细胞凋亡率的变化,免疫荧光技术及流式细胞仪检测核转录因子-κB(NF-κB)的表达情况。结果:ASA在2、4、8Gy三种不同剂量的放射增敏比分别为0.329±0.052、0.404±0.067、0.615±0.033。ASA+照射组的凋亡率[(59.600±1.200)%]较单纯照射组[(39.800±1.124)%]明显增加(P<0.01)。免疫荧光技术及流式细胞仪显示照射后胞核内NF-κB表达增加,而这种增加可被ΑSΑ抑制。结论:ASA可提高鼻咽癌细胞对放射线的敏感性,其机制可能是通过阻止NF-κB的核内转移,继而抑制NF-κB的激活,从而增加放射线诱导的鼻咽癌细胞凋亡。Objective To investigate the effect of aspirin on the radiosensitivity of nasopharyngeal carcinoma cell line SUNE and its mechanism. Methods The proliferation inhibited rate of SUNE cells by aspirin was detected by MTT assay, the apoptosis rate of SUNE cells was measured by flow cytometry, and the expression of nuclear factor-kappa B (NF-KB) was evaluated by immuno-fluorescence and flow cytometry. Results The radiation enhancement ratio of aspirin under the radiation dose of 2 Gy, 4 Gy, and 8 Gy were 0.329 + 0.052, 0.404 ± 0.067, and 0.615 ± 0.033, respectively. The apoptosis rate was increased significantly in radiation (8 Gy) plus aspirin group [(59.600 ± 1.200)% ] than that in radiation alone group[ (39.800 ± 1.124)%] (P 〈 0.01 ). The expression of NF-kB in nucleus was increased after radiation, which could be inhibited by aspirin. Conclusion Aspirin can increase the radiosensitivity of SUNE cells, the possible mechanism may be that aspirin can prohibite the translocation of NF-kB to nuclear, then inhibite the activation of NF-kB, and finally promote apoptosis induced by radiation.
