摘  要:以农杆菌介导的红曲菌T-DNA插入突变库中8 000多株转化子为材料,通过菌落形态观察,获得230株形态变化显著的突变子,并将其分为7类。在此基础上,分析了14株代表菌株产色素、莫纳可啉K(Mo-nacolin K)、桔霉素和淀粉酶的情况。结果表明:所有供试菌株的产色素能力均低于出发菌株M-7,其中806#和2553#的色价均不到M-7的1/10;10株突变菌株产Monacolin K的能力高于M-7,其中3245#产Monacolin K的能力较M-7提高了近10倍,达到679.25μg/g;1822#产桔霉素(干红曲)能力最高,达60.01 ng/g,是M-7的近30倍,而806#和178#产桔霉素能力较低,在试验条件下未检测到;13株突菌株的淀粉酶活高于M-7,1067#最高约为M-7的20倍,达3 227.10 U/g。About 8 000 transformants were obtained by T-DNA insertional library of Monascus tuber mediated by Agrobacterium turnefaciens. 230 of them with significant changes were classified into seven groups according to characteristics of the colony and micro-morphological observation. On this basis, pigment,Monacolin K, citrinin and amylase of the representatives of thel4 mutants were analyzed. The results were as follows, the capacity to produce pigment of all the mutants were poorer than the original strain M-7; 806 ^# and 2553 ^# were the poorest, and the pigment-value of them were less than tenth of M-7. Monacolin K output of 10 mutants was higher than M-7; 3245^# was almost 10 times that of M-7,which amounts to 679.25 μg/g. The citrinin production(dry weight of Hongqu) of 1822^# was the highest,reaching 60. 01 ng/g,nearly 30 times that of M-7; the citrinin production of 178^# and 806^# was not detected under this experimental condition. The activity of amylase of 13 mutants were higher than that of M-7;while the activity of amylase of 1067^# was the higest, up to 3 227. 10 U/g and was 20 times that of M-7.
