摘 要:目的研究LMP1基因C端区在宁波地区鼻咽癌患者中的变异状况,并探讨其在鼻咽癌中所起的作用。方法通过鼻咽拭子的方法采集鼻咽癌患者的病变黏膜上皮及分泌物,采用聚合酶链反应(PCR)扩增鼻咽癌患者鼻咽拭子中的LMP1基因C末端,并对其进行序列分析。结果30例鼻咽癌患者的鼻咽拭子标本中有28例显示出阳性条带,且都存在序列变异,而正常对照标本无一扩增得到,特异性为100.0%。28例阳性标本中有7例标本存在30 bp的缺失突变,缺失率为25.0%。结论宁波地区的鼻咽癌鼻咽拭子中LMP1基因C端高变区30 bp的缺失型约占25.0%,不是主要流行型。Objective To investigate the mutation in carboxy terminal region of Epstein-Barr virus latent membrane proteinl (E-B virus LMP1) gene in patients with nasopharyngeal carcinoma (NPC). Methods Epithelium mucosa and secretion samples were collected by nasopharyngeal swab from 30 patients with nasopharyngeal carcinoma in Ningbo region. LMP1 gene carbexy terminal region was amplified by PCR,and sequence analysis was performed. Results Of the 30 cases from nasopharyngeal patients, 28 were LMP1 positive, and sequence mutation was obtained in all positive cases. Of the 10 cases from normal control,all were LMP1 negative with a specificity of 10G 0%. Of the 28 positive cases,7 were 30 bp deletion mutation with a deletion rate of 25.0%. Conclusion The 30 bp deletion in LMP1 gene may be not the prevalent gene type of nasopharyngeal carcinoma in Ninbo region.
