摘 要:目的研究植物雌激素木黄酮对体外培养的肝星状细胞(HSC-T6)的增殖、活化、胶原合成以及对雌激素受体(ER)表达的影响,探讨木黄酮抑制肝纤维化的作用机制。方法选择细胞系HSC-T6为体外细胞模型,随机分为4组,分别加入不同浓度的木黄酮(0.5、5、50μmol/L),同时设未加任何药物的空白对照组。作用24 h后采用MTT法检测HSC-T6细胞的增殖情况,放射免疫法测定细胞上清液Ⅲ型胶原(PCⅢ)、透明质酸(HA)的含量;免疫组化法观察木黄酮对各组HSC-T6细胞α-平滑肌肌动蛋白(-αSMA)及ER表达的影响。结果木黄酮各剂量组均不同程度地抑制HSC-T6细胞的增殖,降低其-αSMA的表达,减少PCⅢ及HA的分泌,且其抑制作用呈剂量依赖性;给予木黄酮后,HSC-T6细胞的ER表达增强,也呈剂量-效应关系。结论木黄酮可抑制体外培养的HSC-T6细胞的活化与增殖,减少细胞外基质的产生,抑制肝纤维化的形成。其作用机制可能与木黄酮增加HSC的ER表达有关。Objective To investigate the effects of phytoestrogen genistein on the proliferation, collagen synthesis, activation, and the expression of estrogen receptor (ER) in hepatic stellate cell (HSC-T6) in vitro. Methods HSC-T6 cells were randomly divided into genistein group (0.5 μmol/L; 5 μmol/L; 50 μmol/L ) and control group. All groups were simultaneously intervened in 24 hours. Cell proliferation was assessed by MTT colorimetric assay. Hyaluronic acid (HA) and procollagen Type Ⅲ (PC Ⅲ ) were examined by radioimmunoassy (RIA). The expression of a-smooth muscle actin (α-SMA) and ER of HSC-T6 was detected by immunohistochemical method. Results The results showed that the three concentrations as 0.5, 5 and 50 μmol/L of genistein could inhibit the proliferation of HSC-T6, decrease PC Ⅲ, HA and lower expression of alpha-SMA in a dose-dependent manner. The expression of ER in each genistein group was increased compared to that in control group and also in a dose-dependent manner. Oonclusion Genistein has the effect of preventing hepatic fibrosis through inhibiting the proliferation, activation of HSC and collagen synthesis in HSC in vitro, the mechanism of which is partly due to its up-regulation on ER level of HSC-T6 cells.
