摘 要:目的体外实验研究核转运因子2(NTF2)对视网膜新生血管的抑制作用。方法以血管内皮生长因子(VEGF)刺激体外培养人视网膜血管内皮细胞,并分为转染重组腺相关病毒2-NTF2(rAAV2-NTF2)的实验组和转染等量空白rAAV2的对照组,利用流式细胞仪和Boyden小室观察NTF2对激活状态的人视网膜血管内皮细胞增殖和迁徙的作用。结果流式细胞仪检测结果显示实验组在G1期的细胞比例为(49.57±0.43)%,对照组为(27.21±0.86)%,两者差异有统计学意义(P<0.05)。Boyden小室实验检测迁徙细胞数,实验组和对照组分别为(13.8±1.5)和(34.6±2.5)(n=6,P<0.05),说明NTF2能抑制人视网膜血管内皮细胞的增殖和迁徙。结论NTF2对视网膜新生血管具有抑制作用。Objective To study the inhibitory effects of nuclear transport factor 2(NTF2) on retinal neovascularization in vitro. Methods The in vitro cultured human retinal capillary endothelial cells were stimulated with VEGF,and transfected recombinant adenovirus-associated NTF2(rAAV2-NTF2)(experimental group) or blank rAAV2(control group) in vitro. Flow cytometry and Boyden chamber assay were used to detect the effects of NTF2 on the VEGF-stimulated human retinal vascular endothelial cells proliferation and migration,respectively. Results Flow cytometry revealed that the cells in G1 phase in experimental and control groups accounted for (49.57 ± 0.43)% and( 27.21 ± 0.86 )%, respectively, with the difference being significant between them(P〈1:0. 05). Boyden chamber assay demonstrated that the migrating number of cells in experimental and control groups was(13.8± 1.5) and(34.6 ± 2.5) respectively(n= 6, P〈0.05) ,indicating NTF2 could inhibit the proliferation and migration of human human retinal capillary endothelial cells. Conclusion NTF2 can inhibit retinal neovascularization.
