摘 要:[目的]研究不同浓度与不同时间条件下国产吲哚青绿晶状体前囊膜染色对兔角膜内皮活性的影响。[方法]选择健康科研兔18只(36眼),随机分为4组,即A、B、C 3组分别为2.5 mg/mL、5 mg/mL、7.5 mg/mL的国产吲哚青绿溶液组,D组为平衡盐液(BSS)做对照,每组9眼,每组分3亚组分别给予上述溶液作用30、60、90 s,以酶组织化学染色方法测定不同处理的活体兔角膜内皮细胞乳酸脱氢酶(LDH)与还原型辅酶I(NADH)的活性改变。[结果]LDH、NADH活性在A组、B组与D组比较差异均无显著性意义(P>0.05),而C组在30、60、90 s LDH光密度值分别为(186.12±12.12)A、(169.97±11.14)A、(161.82±17.91)A,NADH光密度值分别为(178.86±12.67)A、(165.39±18.79)A、(158.81±17.48)A,LDH、NADH活性在C组与A组、B组、D组比较差异均有显著性意义(P<0.05)。C组组内,酶的活性与药物接触角膜时间呈线性关系,负相关。r=0.45,P<0.05。[结论]2.5 mg/mL与5 mg/mL国产吲哚青绿溶液晶状体前囊膜染色在90 s以内均安全、可靠,对兔角膜内皮细胞活性无影响。而7.5 mg/mL国产吲哚青绿溶液行前囊膜染色在3个时间段对兔角膜内皮活性均产生不同程度影响,酶活性与时间呈负相关。[ Objective] To evaluate the effects of different concentrations and exposure times for the domestic indocyanine green on corneal endothelium in rabbit's eyes. [ Methods ] Thirty- six rabbit eyes were randomly divided into four groups, group A,B and C were 2.5 mg/mL,5 mg/mL,7.5 mg/mL domestic ICG solutions,group D was balanced salt solution(as a control) , then each group was divided into three subgroups with four kinds of solution applied to rabbit eyes 30,60 and 90 sec. The changes in enzymes LDH and NADH activitives after injections were observed. [ Results ] There were no statistical significance for the activities of LDH and NADH among group A, B and D ( P 〉 0.05 ). The activities of LDH in group C directly were ( 186.12 ± 12.12 )A, ( 169.97 ± 11.14 )A, ( 161.82 ± 17.91 ) A in 30,60 and 90 sec, The activities of NADH in group C directly were ( 178.86 ± 12.67 ) A, ( 165.39 ± 18.79) A, ( 158.81 ± 17.48 ) A, The differences of the enzymes activities among the group C and A, B, D were signifficant ( P 〈 0.05 ). The activities of LDH and NADH depend on the ICG exposure time in group C, shown negative correlation between enzymatic activity and exposure times, r = 0.45 ( P 〈 0.05 ). [ Conclusions ] Anterior capsule staining with 2.5 mg/mL,5 mg/mL domestic ICG solution in 90 see are safe, which has no effect on the corneal endothelium activity. While 7.5 mg/mL ICG in three different times had statistical significance on the rabbit's corneal endothelium activity ,which shows negative correlation between enzymes activities and exposure time.
