七氟醚对脑血流量不足的老年大鼠认知功能的影响

摘  要:[目的]观察七氟醚麻醉后对脑血流量不足的老年大鼠认知功能的影响,为临床用药作参考。[方法]24只老年Wistar大鼠随机分为4组,每组6只。分别为对照组(Ⅰ组),2%的七氟醚组(Ⅱ组),3%的七氟醚组(Ⅲ组)和七氟醚+缺血组(Ⅳ组)。各组动物持续分别吸入0%、2%、3%、2%的七氟醚和2Lmin-1O2+6 Lmin-1N2,每天吸入100min,连续吸5d且第Ⅳ组于吸入混合气前1周结扎左侧颈总动脉。用Morris水迷宫测试大鼠的认知功能,每天4次,连续测定5d。水迷宫测试结束后立即处死大鼠,用逆转录-聚合酶链反应方法测定海马NR、NR1和NR2B的mR-NA表达。[结果]①重复测量方差分析结果表明从1d到5d,Ⅰ~Ⅳ组动物的潜伏期和游泳距离有逐日缩短的趋势。到d5各组动物的潜伏期和游泳距离与d1比较差别有统计学意义(P﹤0.05)。②完全随机方差分析结果表明4组NR2B亚单位mRNA表达差别有统计学意义(F=7.79,P﹤0.001)。[结论]七氟醚对老年大鼠术后认知功能呈剂量依赖性抑制作用;脑血流不足影响七氟醚对老年大鼠的认知功能。第Ⅳ组NR2B的mRNA呈现高表达,可能与早前的脑血流不足有关。[Objective] To investigate the effect of sevoflurane on specific cognitive functions in the elderly Cerebral blood flowed deficits rats in order to provide references for clinical drug use. [Methods] Twenty four Wistar rats were randomly divided into 4 groups, including control group (Ⅰ), 2% sevoflurane group (Ⅱ), 3% sevoflurance group (Ⅲ) and sevoflurane-ische-group (Ⅳ). Rats in the four groups were respectively administrated for 100 minutes per day with 0%, 2%, 3% and 2% sevoflurane, and 2L·min-1 O2+6L·min-1 N2, and administrated for five days. Rats in Ⅳ group were ligated left common carotid artery before a week of breathing mixed gas. The cognitive function was tested 4 time a day for 5 days by using Morris water maze. The animals were killed after testing by Morris water maze. mRNA expression of NR, NR1 and NR2B in the hippocampus were determined by reverse transcriptase–polymerase chain reaction (RT-PCR). [Results] ① The repeated analysis of variance showed that the latency period and swimming distance in Ⅰ-Ⅳ group had gradually shorter during the five days, and the latency period and the swimming distance were significantly shorter at the fifth day than those at the first day (P﹤0.05). ② One way analysis of variance showed that mRNA expression of NR2B in group Ⅳ had significant difference among the four groups (F = 7.79, P﹤0.001). [Conclusion] Sevoflurane had dose-dependent inhibited effect on cognitive function of elderly rats. Cerebral blood flow could influence sevoflurane on the cognitive functions in elderly rats. mRNA expression of NR2B in group Ⅳ was higher, and it had association with earlier cerebral blood flow deficits.
