摘 要:目的:研究高浓度铁对大鼠行为学及神经元凋亡的影响,从而进一步探讨其与阿尔采默末病(AD)病因学间的关联。方法:用在大鼠侧脑室埋管、经脑室给药的方法向实验组大鼠脑室注入FeCl3 10μL;向对照组注入人工脑脊液10μL,第5、10天重复给药1次,用Morris水迷宫测定其行为学改变,用Annexin V-FITC染色细胞,通过流式细胞仪测定大鼠早期神经细胞凋亡百分率。结果:行为学实验组大鼠寻找平台时间、距离和均大于对照组;实验组染色阳性细胞百分率高于对照组。结论:本实验证明向脑室内注入铁离子可引起大鼠学习记忆功能障碍的行为学改变及神经元凋亡,这正是AD的特征性临床病理改变,因此推测脑脊液中高浓度铁离子可能是与引起散发性AD的病因有关联。Objective: To examine the behavioral performance and molecular pathological changes induced by higher concentration iron. FeCl3 were injected into intracephalic of rats by embedding a plastic hollow tube into their ventricle of brain. Methods:15 rats were divided into control and experimental groups. Both of the experimental and control rats. A small burr-hole was made in the left/right side of the craium with a dental drill. A plastic hollow tube was embedded in the ventricle of brain. After the surgen ,FeCl3 were injected into ventricle of brain. Of control tats interval 5 days and 3 times injection were carried out. Then the behavioral performance were examined by Morris water maze and the percentage of early apoptosisin of cells load with Annexin-FITC were testded by flow cytomery. Results :There were ignificant differences in the mean escape latencies :sum of escape distance between exprimental and control rats. Percentage of Annexin-FITC loaded cells that are undering apoptosis in animals treated by iron were higher than that of normals. Conclusion:Injecting higher concentration iron into ventrcle induce damage of study and memory and neuron apoptosis which were the clinic and pathological characteristic of AD.
