摘　　要：目的:检测原发性舍格伦综合征(primary Sjgren’s syndrome,pSS)患者血浆和血细胞中IFN-α的表达水平,探讨IFN-α在原发性SS发病中的作用。方法:采用ELISA法和实时定量PCR,对37例原发性SS患者和24例非SS患者进行血浆和血细胞中IFN-α水平检测,比较分析其在两组之间的差异。采用SAS6.12软件包进行t检验和Fisher确切概率检验。结果:SS组中,16例(43.24%)患者外周血浆中IFN-α表达阳性,其余21例(56.76%)为阴性;对照组24例患者外周血浆均为阴性表达,2组有显著差异(P=7.02×10-5)。实时定量PCR法检测发现,SS组的IFN-αmRNA表达量显著高于对照组,2组之间有显著差异(P=0.0124)。结论:IFN-α在原发性舍格伦综合征患者中表达升高。PURPOSE： To investigate the interferon-α level in peripheral blood of patients with primary Sjogren＇s syndrome （pSS）, and explore preliminarily the correlation between pSS and interferon-α. METHODS： ELISA and real- time PCR were used to evaluate the interferon-α level in peripheral blood in 37 patients with pSS and 24 patients with non- pSS. Statistical analysis was performed by Student＇s t test or Fisher＇s exact test using 5AS6.12 software package. RESULTS： The results demonstrated that 43.24% （16/37） patients with pSS had detectable IFN-α concentration in plasma （≥ 12.5pg/mL）, the other 21 patients （56.76%） with pSS had undetectable IFN-α concentration（〈12.5pg/mL）. All 24 controls had undetectable IFN-α expression.There was significant difference between the two groups（P=7.02×10^-5）. Furthermore, the mRNA expression levels of IFN-α gene in peripheral blood cells were over-expressed in patients with pSS （P=0.0124）. CONCLUSIONS： The study reveals that interferon-α level in peripheral blood with pSS patients is overexpressed.
