摘 要:目的:观察犬骨髓基质细胞在体外培养的条件及生物学特性,对其进行鉴定。方法:取犬骨髓分离培养骨髓基质细胞(BMSC)传代后,倒置相差显微镜观察其形态,免疫组化方法加以鉴定,生化检测Vonkossa染色鉴定其成骨能力。结果:原代培养20d后可分离得到骨髓基质细胞BMSC,生长形态相对稳定,具有成骨能力。结论:BM-SC具有贴壁生长特征,易分离扩增,增殖速度快,诱导分化具有成骨能力,可用于骨缺损的修复治疗。Objective:To observe in vitro culturing condition and biological chatacteristics of bone marrow stromal cells(BMSC) of dogs and identify the cells. Methods:Bone marrow of dogs were separated and the BMSC were cultured. The morphology of the cells was studied with a phase contrast microscope and the living characteristics after subculture of cells were observed. Result:BMSC could be observed after 20 days primary culture. The living biological chatacteristics were quite stable,and owned the bone formation activity. Conclusion:BMSC can be easily separated and proliferated fastly,which might be used to repair bone defect.
