摘 要:目的:目前LCRG1基因转录调控机制不清,现拟研究Sp1和Egr-1对人LCRG1基因启动子的转录调节。方法:利用MatInspector软件分析LCRG1基因启动子区域内潜在的转录因子结合位点,Sp1、wtEgr-1、mtEgr-1真核表达质粒与LCRG1启动子重组质粒的共转染实验,分析其对LCRG1启动子活性的影响。结果:生物信息学提示LCRG1基因启动子区域存在Sp1和Egr-1等位点,外源性突变型转录因子Egr-1能上调LCRG1基因启动子的活性。结论:突变型转录因子Egr-1可能参与该基因的表达调控。Objective:The regulatory mechanisms of LCRG1 gene expression is undear,this study was to investigate the transcription regulation of the promoter of human LCRG1 gene by transcription factors Sp1 and Egr-1.Methods:Analysis of putative binding sites of transcription factors in the promoter of human LCRG1 gene by online program MatInspector,Co-transfection with the expression plasmids of the known transcription factors such as Sp1、wtEgr-1、mtEgr-1with the LCRG1 reporter construct analyze potent transcription factor for enhancement of the promoter activity.Results:SP1 and Egr-1 transcription factor binding site sites were predicted in the region by bioinformatics analysis,mtEgr-1 can enhance the promoter activity.Conclusion:mtEgr-1 may be involved in the regulatory mechanisms of LCRG1 gene expression
