摘  要:目的:腹主动脉瘤(AAA)是一类死亡率极高的主动脉疾病。为研究AAA的发病机制及治疗措施,必须建立合适的病理模型。文中研究建立简便、迅速、稳定的Wistar大鼠AAA模型。方法:10周龄雄性Wistar大鼠30只,随机分成实验组及对照组(各15只),实验组采用缩短弹力酶灌注时间的改进建模方法,对照组采用传统建模方法。14 d后计算2组动物的存活率、腹主动脉瘤的形成率,制作腹主动脉石蜡及电镜切片,观察其形态学变化。结果:对照组大鼠手术存活率为60%,存活大鼠AAA形成率为89%;实验组大鼠存活率为87%,存活大鼠AAA形成率为85%;两组大鼠均可见腹主动脉明显扩张,动脉壁变硬,部分区域可见内膜撕裂,中膜弹力纤维减少或消失,外膜炎性细胞浸润明显。结论:改进后的30 min弹力酶灌注法可用于AAA大鼠模型的制作,动物成活率及AAA形成率均较高。是一种简便、迅速、稳定的腹主动脉瘤Wistar大鼠建模方法。Objective: Abdominal aortic aneurysm(AAA) is a kind of abdominal aortic disorders with very high mortality.This paper reports the innovated methods for a simple,quick,stable and high-successed-rate model of abdominal aortic aneurysms(AAA).Methods: Wistar rats were divided into control group and improving method group,and AAA models were made with traditional and modified method respectively.The two methods were compared with formation and mortality.Results: Both of the methods may induce AAA.The mortality of control group rats was 40 percents,modified method group was 14 percent.Light microscopy showed that the development of AAA in vehicle-treated rat was accompanied by marked thickening of the aortic wall,a dense transmural inflammatory response,and complete destruction of the elastic media.Conclusion: Modified method is a simple,quick,stable and high-success-rate model of abdominal aortic aneurysms.
