摘  要:目的:建立大鼠甲基苯丙胺(methaphetamine,MA)慢性中毒模型,探讨MA长期应用对脑纹状体区的损伤作用。方法:Wistar大鼠30只,雄性,体重180-220 g,随机分为对照组(A)和实验组(B)。B组从小剂量开始腹腔注射MA,逐渐加量,共用4周;A组腹腔注射等量生理氯化钠溶液。电子体温计监测体温变化;TUNEL法观察纹状体内神经元凋亡情况;高效液相色谱检测纹状体内多巴胺(dopamine,DA)含量。结果:MA引起大鼠行为活动明显增多;纹状体区神经细胞凋亡数量显著增多及DA含量明显下降。结论:MA慢性中毒对大鼠脑组织纹状体有明显的毒性损伤,可引起大鼠行为改变,纹状体神经元大量凋亡及DA含量显著下降。Objective: To establish a rat model of chronic toxicity of methamphetamine (MA) and to explore MA-induced chronic neurotoxicity on striatum. Methods: Thirty male rats, weighting 180 -220 g, were randomly divided into 2 groups, and treated with either gradually increasing doses of MA or saline (control) for 4 weeks. The body temperature was monitored; the changes in the behaviors were observed and graded. Neuron apoptosis was detected by TUNEL straining. The contents of the dopamine (DA) were detected with HPLC. Results: The locomotor activities increased remarkably and the number of apoptotic neurons in the striatum increased obviously after MA administration. The contents of dopamine in the striatum decreased obviously. Conclusion: Chronic application of MA induces obvious damage of the striatum, dysfunction of behaviors, neuron apoptosis and decrease in the contents of striatum dopamine.
