胃癌组织半乳糖凝集素1表达的意义

摘　　要：目的:研究半乳糖凝集素1(galectin-1)在胃癌组织中的表达,分析其在胃癌生长、侵袭中的作用.方法:外科手术切除的胃癌组织及胃切缘组织标本30例,采用RT-PCR方法从mRNA表达水平检测胃癌组织及远癌切端胃组织中Ga-lectin-1的表达情况,并用免疫组织化学SABC法检测Galec-tin-1蛋白表达及其微血管密度(MVD).结果:胃癌组织Ga-lectin-1 mRNA及蛋白表达水平、MVD均分别显著高于远癌切端胃组织,差异均具有统计学意义(P<0.05).胃癌组织Ga-lectin-1 mRNA和蛋白表达水平在不同浸润深度、TNM分期中具有显著差异(P<0.05).MVD在不同浸润深度、TNM分期及有无淋巴结转移的胃癌组织中具有显著差异(P<0.05).胃癌组织Galectin-1 mRNA和蛋白表达水平与胃癌MVD具有显著的正相关(P<0.05).结论:Galectin-1参与胃癌生长和侵袭,可作为诊断、判断进展程度及预后的指标.AIM：To study the expression of Galectin-1 in gastric cancer and to analyze its correlation with the invasion of gastric cancer. METHODS：Cancer samples and surgical margin of stomach were collected from 30 patients who had undergone surgery in General Hospital of Tianjin Medical University. Real-time quantitative RT-PCR and immunohistochemistry were used to detect the expression of Galectin-1 in these samples and the microvessel density （MVD） of these samples were detected by immunohistochemistry. RESULTS：The expression of Galectin-1 mRNA and protein in cancer and the MVD of cancer were significantly higher than those in tissues of surgical margin of stomach （P〈0.05）. Significant correlation was found between the expression of Galectin-1 mRNA and protein in cancer and the TNM stage and depth of primary tumor invasion （P〈0.05）. The MVD of cancer was statistically correlated with the depth of primary tumor invasion,TNM stage and lymph node metastasis （P〈0.05）. In gastric cancer,there was a positive correlation between MVD and the expression of Galectin-1 mRNA and protein （P〈0.05）. CONCLUSION：Galectin-1 plays a crucial role in gastric cancer＇s proliferation and invasion. It could be a valuable index for clinical diagnosis and malignant progression of gastric cancer.
