摘 要:目的探讨灵芝多糖(GLP)和核心蛋白聚糖(DCN)基因联合使用对S180荷瘤小鼠免疫功能影响及其抗肿瘤作用。方法将32只小鼠,建立S180实体移植瘤模型,随机分为生理盐水组(NS组)、灵芝组(GLP组)、转基因治疗组(DCN组)和灵芝联合转基因治疗组(DCN+GLP组)。测量小鼠肿瘤体积,并采用MTT法检测T淋巴细胞转化能力,ELISA测定血清VEGF的变化,流式细胞检测CD4+CD2+5调节性T细胞占CD4+T细胞的比例。结果各治疗组平均肿瘤体积增长延缓,淋巴细胞增殖率显著增加(P<0.05),同时VEGF表达明显下调,脾脏CD+4CD2+5调节性T细胞占CD4+T细胞的比例明显降低。结论灵芝多糖和核心蛋白聚糖DCN联合使用能够显著提高荷瘤小鼠免疫系统的活性,抑制肿瘤生长。Objective To investigate the impacts of the combined therapy of ganoderma lucidum polysaccharide(GLP)and decorin(DCN)gene on immunological function and its anti-tumor effect in mice bearing S180.Methods S180 solid transplanted carcinoma models were established among 32 mice,which were randomized into normal saline group(NS group),GLP group,DCN group and DCN+GLP group.The volume of tumors was measured and MTT method was adopted to test the transformation of T lymphocytes.ELISA was used to test the changes in serum VEGF. Flow cytometry was used to test the percentage of CD4^+ CD25^+ regulatory T cells in CD4^+ T cells. Results It was found in every treatment group that the growth of tumor was slow down averagely in volume,the proliferative rate of lymphocytes was increased significantly ( P 〈 0.05 ), VEGF expression was downgraded apparently and the percentage of CD4^+ CD25^+ regulatory T cells in CD4^+ T cells was reduced obviously. Conclusion The combination of GLP and DCN gene can improve significantly the activity of immune system of tumor bearing mice and suppress tumor growth.
