摘　　要：目的研究整合素αvβ3对大鼠C6脑胶质瘤细胞增殖的影响,为以整合素αvβ3为靶点治疗脑胶质瘤提供实验依据。方法采用MTT比色法和SABC免疫组化检测法,观察不同浓度的整合素αvβ3抗体对体外培养的大鼠C6脑胶质瘤细胞增殖的抑制作用和对增殖细胞核抗原(PCNA)表达的影响。结果整合素αvβ3抗体对体外培养的大鼠C6脑胶质瘤细胞的增殖和PCNA表达有明显的抑制作用,其作用表现出明显的量效关系(P<0.05)。结论整合素αvβ3对脑胶质瘤细胞的恶性增殖具有促进作用,而整合素αvβ3抗体对其有明显抑制作用,为抗整合素αvβ3治疗脑胶质瘤提供了实验依据和理论依据。Objective To investigate the effect of integrin αvβ3 on cell proliferation of rat C6 glioma cells in vitro, so as to provide more experimental bases for brain glioma treatment by anti-αvβ3 integrin. Methods The effect of various concentration of integrin αvβ3 monoelonal antibody on cell proliferation and PCNA expression of rat C6 glioma cells in vitro was detected by MTr colorimetic method and SABC immunohistochemistry for observing expression of proliferation cell nuclear antigen （PCNA）. Results Integrin αvβ3 monoclonal antibody markedly inhibited cell proliferation and PCNA expression of rat C6 glioma cells in a dose-depended manner （ P 〈 0. 05 ）. Conclusions Integrin αvβ3 can positively regulate the cell malignant proliferation of glioma,and integrin αvβ3 antibody can inhibit it. This result provides more experimental and theoretical bases for glioma treatment by anti-αvβ3 integrin.
