摘　　要：目的:探讨二烯丙基二硫(diallyl disulfide,DADS)诱导人结肠癌SW480细胞凋亡的作用。方法:采用MTT、细胞计数法,观察不同浓度和作用时间DADS对SW480细胞的生长抑制作用;施用流式细胞术检测不同浓度DADS对SW480细胞的凋亡率。结果:MTT法显示,DADS能明显抑制SW480细胞生长增殖,呈浓度和时间依赖性,细胞生长增殖速度明显减慢。细胞计数表明,当DADS浓度增加时,SW480细胞群体倍增时间延长,与对照组比较,差异有显著性意义(P<0.05)。流式细胞仪分析显示,随着DADS处理浓度增加,使SW480细胞凋亡率逐渐增高,差异有显著性意义(P<0.05)。结论:DADS具有诱导人结肠癌SW480细胞凋亡的作用。Objective：To investigate whether DADS induces apoptosis in SW480 cells. Methods：The growth inhibition effect of DADS of different concentration on SW480 cell line was measured by MTr assay and cell counting. Apoptosis rate was analyzed by flow cytometry. Results：The MTr assay showed that D ADS inhibited growth of SW480 cells significantly with dose dependent. Adding 25μg/ ml,35μg/ml,50μg/ml and 70μg/ml DADS for 48 hours, SW480 cell growth were suppressed by 18.67% ,33.02% ,49.12% ,66. 86%, respectively. There were significant difference between the treated and controlled cases （ P 〈0.05 ）. Cell counting results showed that average doubling time were retarded from 34.68h in normal cultured SW480 cells to 96.33h in 70μg/ml DADS treated SW480 cells （ P 〈0.05）. Flow eytometry analysis revealed that SW480 cells treated with 25μg/ml,35vg/ml,50μg/ml and 70μg/ml DADS increased the apoptosis rate. Conclusion： Diallyl disulfide could significantly induce apoptosis in SW480 cells.
