摘  要:目的:探讨不同转移能力的肺癌细胞株中miRNA表达差异,并分析miRNA在肺癌转移过程中的功能及意义。方法:利用非编码小RNA芯片技术,检测高转移肺癌细胞株(PGBE1)与低转移肺癌细胞株(PGLH7)间miRNA的表达差异,并通过qPCR鉴定。结果:非编码小RNA芯片分析显示,在不同转移能力肺癌细胞中得到4条差异表达的miRNA,分别为hsa-mir-34b、hsa-mir-23a、hsa-mir-23b和hsa-mir-199a-5p。实时定量PCR对差异的miRNA进行鉴定,4条差异的miRNA在低转移细胞株(PGLH7)中的丰度均高于其在高转移细胞株(PGBE1)中的丰度,P<0.05,与芯片结果吻合。结论:4条miRNA可能与肺癌转移相关,对miRNA功能探讨将有助于研究肺癌转移的分子机制。OBJECTIVE: To identify the specific expression profile of microRNA (miRNA) in the different cell line (PGBE1 and PGLH7) and discuss the potential biological function of the specific miRNAs. METHODS: the expression profile of miRNA in the different transferring cell lines were checked by the non coding RNA chip. The specific expressed miRNAs were identified by quantitive PCR. RESULTS: Four differentially expressed miRNA (hsa-mir-34b, hsa-mir-23a, hsa-mir-23b and hsa-mir-199a-5p) were indentified by quantitive PCR. The expression level in PGLH7 was higher than the level in PGBE1, P 〈 0.05. CONCLUSION: Four miRNAs were identified to related with the transferring ability of lung cancer.
