摘 要:目的血啉甲醚(HMME)和癌光啉(PsD-007)在鼻咽癌动物模型中的稳态和瞬态荧光光谱特性。方法将接种CNE-2鼻咽癌细胞的裸鼠30只随机分为10组,尾静脉注射光敏剂20mg/kg,经过3、6、24、36和48h代谢后,运用FLS920荧光分光光度计测量稳态荧光光谱。经过光谱分析和处理,得到不同代谢时间下肿瘤组织和正常组织中的光敏剂含量。用405nm超短脉冲激光器作为激发光源,探测肿瘤组织在625nm处的荧光衰减曲线,通过双指数拟合,计算不同代谢时间肿瘤组织的荧光平均寿命。结果肿瘤和正常组织的荧光发射特征峰均在625nm和695nm,HMME和PsD-007的最佳光动力诊断时间分别为3和6h。不同代谢时间的肿瘤组织荧光平均寿命之间没有显著差异。结论通过检测血啉甲醚和癌光啉在鼻咽癌动物模型中的荧光特性,可以实时反映光敏剂在肿瘤和正常组织中的代谢情况,这对于确定最佳的光动力诊断时间具有指导意义。瞬态光谱的检测可为光动力诊断提供更加全面的信息。Objective To study the steady-state and time-resolved fluorescence properties of HMME and PsD-007 in nude mice bearing nasopha- ryngeal carcinoma (NPC) , respectively. Methods The mice were randomly divided into l0 groups (3 in each group) according to the type of photosensitizers (PS) and detec- tion times (3, 6, 24, 36 and 48 h ) , and the mice with more than 5 mm diameter tumors were given an intravenous injection of 20 mg/kg of HMME or PsD-007. The FLS920 spectrofluorophotometer was used to collect the steady-state spectra, and the amount of photosensitizers in tumor and normal tissue at different detection time was determined with spcetroscopic analytical method. 405 nm Ssuper-short pulse laser was used as excitation light source for measurement of time-resolved fluorescence spectra. The two-eompoent lifetimes of NPC were calculated from the measured time-resolved fluorescence spectra.Results The emission wavelengths of photosensitizers in tissues were located at 625 and 695 nm under excitation at 488 or 514. 5 nm. The data indieated that the optimal diagnostic times were 3 and 6 hours for HMME and PsD-007 after injection, respectively. There was no significant difference for fluoreseence lifetimes for NPC at different measured time. Conclusions The metabolism condition of photosensitizers in tumor and normal tissue can be quantified by fluorescence method and to further determine the optimal diagnostic time.
