摘 要:目的观察高糖培养对大鼠近端肾小管上皮细胞肥大的影响。方法SD大鼠,麻醉下,无菌分离单根近球小管并培养,以免疫细胞化学方法鉴定体外培养的肾小管上皮细胞。不同浓度糖(10mM,20mM,30mM)培养肾小管上皮细胞,72小时后检测细胞体积、3H-亮氨酸掺入量以及蛋白质含量,以观察细胞肥大情况。结果高糖(20mM,30mM)培养72小时,导致肾小管上皮细胞体积明显增大,3H-亮氨酸掺入量及细胞内蛋白质含量明显增加。结论高糖培养可诱导肾小管上皮细胞肥大。Objective To investigate the effect of high glucose on the hypertrophy of renal proximal tubular epithelial cells in rats. Methods Renal proximal tubular epithelial cells were gained by microdissection and cultured under different concentration of glucose. The cell types were idenfied by immunocytochemistry. Cell size , ^3H - leucine incorporation, and cellular protein content were detected to observe the changes after renal proximal tubular epithelial ceils were incubated with different concentration of glucose (10 mM, 20 mM, 30mM) for 72 hours.Results The renal proximal tubular epithelial cells were cultured with different concentration of glucose (10 mM,20 mM,30mM) .Following the increase of glcose concentration,cell size, ^3H - leucine incorporation and cellular protein content increased significantly.Conclusions The result of the present investigation suggests that high glucose induces hypertrophy of renal proximal tubular epithelial cells.
