摘 要:目的探讨环境因素(如饮食)与遗传基因的相互作用对个体血脂性状的影响。方法以高脂乳剂喂养120只小鼠4w后,采用PCR-SSCP(聚合酶链反应单链构象多态性)方法检测小鼠脂蛋白脂酶(LPL)基因型对其血脂水平和部分器官性状的影响。结果高脂乳剂喂养4w后,120只小鼠中98只患有高脂血症,22只未患。在对所有小鼠基因组DNA进行PCR扩增和单链构象多态分析,发现在所扩增的220bp目的片段的14355处(GenBank accession No.NT039462)存在一个G/A的突变。相关性分析表明,BB基因型(A/A)小鼠的TC、TG、LDL、心/体重(H/W)、肝/体重(L/W)明显高于AA基因型(G/G)小鼠,差异显著(P<0.05);而AA基因型小鼠的HDL高于AB和BB基因型(P<0.05)。结论不同LPL基因型小鼠对高脂乳剂的敏感性存在一定的差异。Objective To investigate the effect of diet and gene on blood-fat trait of the individual. Method One hundred and twenty mice were fed with high fat emulsion for 4 w, then the genotypes of LPL were analyzed by PCR-SSCP to investigate their effect on blood-fat traits and some organ performance of mice. Result After fed with high fat emulsion for 4 w, 98 mice suffered from hyperlipidemia, while 22 mice did not. And a G/A mutation was found in 220 targeted fragments we amplified at 14355 site, and association analysis showed that the mice with BB genotype had higher TC, TG, LDL, H/W, L/W level than those of the mice with AA genotype (P〈 0.05 ) , and the mice with AA genotype had higher HDL level than those of the mice with AB and BB genotypes (P〈0.05) . Conclusion Different LPL genotypes show different sensitivity to high fat emulsion in mice.
