摘  要:目的研究新疆哈萨克族食管癌中Cdc42基因启动子区甲基化状态与其mRNA表达改变的关系。方法用RT-PCR方法测定mRNA表达,甲基化特异PCR方法检测DNA启动子区甲基化状态。结果在20例癌组织中,有16例表达阳性,占80%,20例癌旁组织中有13例表达,占65%,其中癌组织表达量明显高于癌旁组织的有10例,占50%,癌旁组织表达量高于癌组织的有5例,占25%。肿瘤组织和正常组织该基因启动子区见甲基化条带,未见非甲基化条带。结论Cdc42基因在肿瘤组织和正常组织均表现为高甲基化状态,新疆哈萨克族食管癌中Cdc42基因mRNA表达增强可能与该基因的甲基化状态没有直接关系,另存在其他机制,有待进一步研究。Objective To study the expression of Cdc42 and promoter region methylation in Hazak esophageal cancer. Methods Cdc42 expression and Cdc42 promoter region methylation, were detected in tissues of 20 cases with esophageal cancer and tumor -adjacent normal tissues by reverse transcription polymerase chain reaction ( RT - PCR ) and methylation - specific PCR ( MSP). Results The overall positive rate of Cdc42 gene expression was 80% in the tissues of 20 cases with esophageal cancer, and 65% in tumor- adjacent normal tissues. Promoter region methylation was observed in esophageal cent normal tissues. Conclusions It is suggested that the cell apoptosis and cancer tissues and tumor - adjacell cycle changes resulted from the abnormal expression of Cdc42 gene affect the occurrence of Hazak esophageal cancer, but the promoter region methylation is not relative with the gene expression of Cdc42. So, the promoter region methylation is less important for the gene expression in Cdc42 gene than in others.
