摘  要:[目的]观察不同浓度雌激素(17β-estradiol,17β-E2)对体外培养人牙周膜细胞(human perio dontal ligament cells,HPDLCs)增殖和骨保护素(osteoprotegerin,OPG)表达的影响,探讨雌激素对牙周组织改建的作用。[方法]体外培养HPDLCs,MTT法测定不同浓度17β-E2(0、10-5、10-6、10-7mol/L)作用下HPDLCs增殖水平;ELISA法和RT-PCR法确定OPG分泌量及mRNA表达。[结果]MTT结果显示加入不同浓度17β-E2对HPDLCs增殖无显著影响(P﹥0.05);各浓度组作用于HPDLCs均可促进OPG蛋白分泌和mRNA表达,同一时间点与0mol/L对照组比较,10-7、10-6mol/L组差异有统计学意义(P﹤0.05)。[结论]17β-estradiol不影响牙周膜细胞增殖;生理浓度17β—estradiol可以促进HPDLCs的OPG表达,有利于牙槽骨保护。[Objective] To investigate the effects of estrogen (17β-estradiol, 17β-E2) on proliferation of cultured hu-man periodontal ligament cells and osteoprotegerin expression, and explore the role of estrogen on the periodontal remodeling. [Methods] Human periodontal ligament cells were cultured in vitro and its ability of proliferation was measured by MTT assay under the effect of 17β-estradiol at different concentrations (0, 10-5, 10-6, 10-7mol / L); secretion levels of the OPG mRNA expression were determined by ELISA and RT-PCR methods. [Results] MTT assay showed that treatment with different concentrations of 17β-E2 had no significant effects on the proliferation of human periodontal ligament cells (P﹥0.05); each of 17β-E2 groups act on HPDLCs enhanced the OPG protein secretion and mRNA expression. As compared with 0 mol / L control group at same time, the difference was significant (P﹤0.05) in the 10-7, 10-6mol / L groups. [Conclusion] 17β-estradiol does not affect proliferation of human periodontal ligament cells and can promote the OPG expression in HPDLCs at physiological concentration, and it is beneficial to preserve the alveolar bone.
