穴位埋线对拟阿尔茨海默病大鼠海马超微结构和胶质纤维酸性蛋白的影响

摘  要:目的:探讨穴位埋线对拟阿尔茨海默病(Alzheimer’s Disease,AD)大鼠海马CA1区超微结构和胶质纤维酸性蛋白(GFAP)的影响。方法:采用冈田酸(Okadaic acid,OA)海马CA1区多次微量注射建立拟AD模型大鼠,应用肾腧(双侧)、大椎穴位埋线治疗;用Morris水迷宫实验检测动物的学习记忆能力,电镜观察海马神经元超微结构,免疫组织化学方法观察GFAP的表达。结果:(1)模型组大鼠平均逃避潜伏期明显延长,第Ⅲ象限活动时间明显缩短及穿越站台次数明显减少,海马CA1区神经元超微结构有典型的凋亡改变,GFAP免疫反应阳性细胞明显增多;(2)穴位埋线组大鼠平均逃避潜伏期明显缩短,第Ⅲ象限活动时间明显延长及穿越站台次数明显增多,海马CA1区神经元凋亡现象改善,GFAP免疫反应阳性细胞明显减少。结论:穴位埋线可能是通过改善AD模型大鼠海马的超微结构和降低GFAP表达的机制而提高AD大鼠的学习记忆能力。Objective: To study the effects of Catgut implantation at acupoints on ultrastructures and glial fibrillary acidic protein(GFAP) of hippocampus neurou in Alzheimer disease(AD)-like model rats.Method: Okadaic acid(OA) was injected into hippocampal CA1 region(HCR) of rats with microdosis repeatedly to establish Alzheimer disease-like model.Catgut was implanted in Shenshu and Dazhui acupucture points once every seven days.The learning and memory capabilities of rats were assessed through Morris Water Maze behavioral test.Ultrastructure of HCR was observed with electron miscroscope and expression of GFAP was examined with immunohistochemistry method.Results:(1) The average escape latency of the model rats was prolonged in the place navigation test and their swimming time in the third quadrant shortened in the spatial probe test,and the number of traversing platform decreased after the platform was removed.Typical apoptosis of neurons were observed in hippocampus,and the expression of GFAP in the HCR increased;(2) The average escape latency of the catgut implanted rats was shortened in the place navigation test,and their swimming time in the third quadrant was prolonged in spatial probe test,and the number of traversing platform increased after the platform was removed.Apoptosis of neurons were lightened in HCR.The expressions of GFAP in HCR decreased.Conclusion: The mechanism of that catgut implantation at acupoint of Shenshu and Dazhui improve the learning and memory capabilities of the model rats might relate to the improvement of the ultrastructure and decrease of GFAP expression level in HCR.
