摘 要:[目的]对红色素生产菌粘质沙雷氏菌的培养基进行优化。[方法]以粘质沙雷氏菌RZ-21为供试菌种,研究培养基中碳源、氮源种类及用量对菌体生物量和红色素产量的影响。[结果]以黄豆粉为碳源时,菌体生物量和红色素产量分别为15.994和1.551 g/L,分别为基础培养基(牛肉膏为碳源)的1.72和1.91倍;以酪素和蚕蛹粉为氮源时,菌体生物量分别为18.755和19.873 g/L,分别为基础培养基(以蛋白胨为氮源)的2.04和2.16倍,红色素产量分别为2.025和1.744 g/L,分别为基础培养基的2.51和2.17倍;全波长扫描和质谱分析结果显示,该红色素为灵菌红素。[结论]粘质沙雷氏菌发酵生产红色素的最佳培养基为:黄豆粉0.5%,酪素1%,蚕蛹粉1%,K2HPO40.2%,NaCl 0.5%,此条件下,菌体生物量和色素产量分别为26.587和2.642 g/L。[Objective] The aim was to optimize the medium for red pigment producing strain Serratia marcescens RZ-21. [Method] With S. marcescens RZ-21 as the tested strain, the effects of different kinds of carbon resource and nitrogen resource in medium on the biomass yield and red pigment yield were studied. [Result] When carbon resource was soybean flour, the biomass yield and red pigment yield were 15.994 and 1.551 g/L resp. ,being 1.72 and 1.91 times of those in basis medium (beef extract as carbon resource ).When nitrogen resource were casein and pupa powder, the the biomass yield were 18.755 and 19.873 g/L resp, being 2.04 and 2.16 times of those in basis medium (peptone as nitrogen resource) resp. ,and the red pigment yields were 2.025 and 1.744 g/L resp. , being 2.51 and 2.17 times of those in basis medium-The result of full spectrum scan and mass spectrum analysis showed that the red pigment was modigiosin. [Conclusion] The optimal medium for S. marcescens producing red pigment was that 0.5% soybean powder,1%casein,1% pupa powder,0.2% K21-1PO4,0.5%NaCl.Under that condition, the biomass yield and red pigment yield were 26.587 and 2.642 g/L resp.
