摘 要:目的将小鼠骨髓来源的DC细胞和小鼠胃癌细胞MFC进行融合,同时以CD44v5进行修饰,研究CD44v5修饰DC融合瘤苗抗肿瘤作用及其作用机制,为肿瘤免疫治疗提供实验依据及理论基础。方法①在PEG的作用下,DC细胞和MFC细胞进行融合,利用HAT/HT系统筛选出纯净融合细胞株。在Lipofectamine 2000的介导下,将pUC19-CD44v5转染到肿瘤融合细胞中表达;②荷瘤小鼠接种融合瘤苗,观测小鼠的生存期及种植瘤组织病理形态学变化情况。结果免疫组小鼠生存期延长,种植瘤组织病理形态明显改变。结论CD44v5修饰的DC融合细胞可有效诱导T淋巴细胞产生强大抗胃癌作用。Objective To study the antitumor effect and mechanism of fusion hybrid of dendritic cells and gastric cancer cell MFC expressing CD44v5, the dendritic cells and gastric cancer cell MFC derived from mice were fused, and modified with CD44v5, it will provide the experimental and theoretical foundation on the immunotherapy for tumor patients. Methods (1)DCs and MFC cells were fused by polyethylene glycal(PEG) , and the completely fused ones were screened by HAT/HT system. The recombinant pUC19-CD44v5 vector was transfected into the fused cells (hybridoma cell) by lipofectamine 2000, and to express;(2)Fused cells were implaned to bearing-cancer mice. The life span of mice and the pathomorphology of the growth tumor were studied. Result The life span of immune group was extended, and the pathomorphology of the growth tumor was obviously changed. Concluslon The fused cells of DC expressing CD44v5 can induce strong antigastric-cancer effect of T lymphocyte.
