摘  要:目的:探讨非受体酪氨酸激酶c-Abl/Arg对细胞周期的影响。方法:用si-STRIKE-c-Abl/Arg siRNA表达载体转染乳腺癌细胞MCF-7,筛选具有潮霉素(hygromycin)抗性的c-Abl/Arg双敲低的稳定细胞系(c-Abl/Arg-DKD),用流式细胞仪检测其细胞周期。结果:筛选获得的c-Abl/Arg-DKD稳定细胞系中c-Abl/Arg的表达被有效抑制,且在S/G2期产生显著的细胞周期阻滞效应。结论:c-Abl非受体酪氨酸激酶在S/G2周期转换过程中发挥调节作用。Objective:To establish the c-Abl/Arg double knock-down stable cell line and investigate the effect of c-Abl/ Arg RNA on MCF-7 cell cycle progression. Methods:si-STRIKE-c-Abl/Arg Plasmid, which was directed against c-Abl/ Arg expression, was transfected into MCF-7 cells. The stable siRNA-mediated c-Abl/Arg knockdown cell lines selected by hygromycin were identified by Western blot. Cell cycle progression was detected by flow cytometry. Results:More than 90% of the decrease in c-Abl expression in c-Abl/Arg-DKD stable cell lines resulted in remarkable S/G2 cell cycle arrest. Conclusion :The results demonstrated that c-Abl tyrosine kinase is involved in the regulation of S/G2 cell cycle transition.
