摘 要:目的检测Dickkopf1(DKK1)在肝癌组织中的表达。方法采用四种不同方法检测DKK1在肝癌组织和癌旁肝组织的表达。结果半定量逆转录聚合酶链反应(RT-PCR)方法检测显示,DKK1在4/5肝癌中高表达;适时-PCR(Real-time PCR)方法检测显示,DKK1在8/14肝癌中高表达;Western blot方法检测显示,DKK1在2/4肝癌中高表达;免疫组化方法检测显示,DKK1在46/79肝癌中高表达,但与AFP表达水平、分化程度、TNM分期等临床病理特征无关。四种方法检测结果均显示DKK1在肝癌组织的表达显著高于癌旁肝组织。结论DKK1在肝癌组织表达高于癌旁肝组织,可能与肝癌发生、发展相关。Objective To explore the expression of Dickkopf1(DKK1) in human hepatocellular carcinomas (HCCs). Methods The expression of DKK1 in HCC samples and tumor-adjacent tissues was detected using four different techniques. Results DKK1 was overexpressed in 4 out of 5 HCC tissues detected by semi-quantitative reverse transcription polymerase chain reaction, which was in 8 out of 14 HCC tissues detected by real-time PCR, and in 2 out of 4 HCC tissues detected by Western blotting. The immunohistochemistry analysis of formalin-fixed wax-embedded sections demonstrated that the overexpression of DKK1 was found in 46 out of 79 HCC patients,which was not correlated with clinicopathologie feature. The expression of DKK1 was significantly higher in HCC tissues than that in tumor-adjacent tissues detected by the four techniques. Conclusion The expression of DKK1 was significantly higher in HCC tissues than that in tumor-adjacent tissues,which might be related to carcinogenesis and development of HCCs.
